Özet
Amaç: Mekanosensitif Piezo 1 ve Piezo 2 kanallarının endometrium kanseri ve over kanserindeki relatif mRNA ekspresyonlarının belirlenmesi; endometrium kanseri ve over kanseri teşhisinde potansiyel bir belirteç olup olmadıklarını belirlemektir. Gereç ve Yöntem: Atatürk Üniversitesi Eğitim ve Araştırma Hastanesi'nde opere edilmiş over kanseri ve endometrium kanseri hastalarına ait patoloji bloklarından ve sağlıklı over ve endometrium dokusu içeren bloklardan kesitler alındı. Alınan kesitler patoloji bloklarından RNA eldesine uygun bir kit kullanılarak total RNA izolasyonu yapıldı. Takiben elde edilen RNA'lardan cDNA sentezi yapıldı. Piezo 1 ve Piezo 2 gen ekspresyonları Real Time PCR (qPCR) yöntemi ile belirlendi. Bulgular: Endometrium kanseri dokusunda hem Piezo 1'in hem de Piezo 2'nin normal endometrium dokusuna göre anlamlı derecede ekspresyon artışı gösterdiği gözlemlendi. Tümör hacmi, lenfovasküler invazyon ve p53 ekspresyonu ile Piezo 1 ekspresyonları arasında yapılan pearson korelasyon testlerine göre istatistiksel olarak anlamlı ilişki bulunmaktaydı. Over kanseri dokusunda hem Piezo 1'in hem de Piezo 2'nin normal over dokusuna göre anlamlı derecede ekspresyon artışı gösterdiği gözlemlendi. Yapılan Pearson korelasypn testlerine göre istatistiksel olarak anlamlı çıkan bir patoloji özelliği olmamasına rağmen preoperatif Piezo 1 ve Piezo 2 ekspresyonları bazı patolojik değerler ile zayıf-orta ve orta derecede korelasyon vermiştir. Sonuç: Piezo 1 ve Piezo 2 ekspresyonlarının endometrium kanseri ve over kanserinde sağlıklı dokulara göre ekspresyon artışı ile beraber çeşitli klinik belirteç, klinik durumlar ve patolojik bazı özelliklerle de korelasyon içinde olduğu görülmüştür. Piezo 1 ve Piezo 2 endometrium kanseri ve over kanseri için henüz çok erken olsa da bir biyobelirteç olmaya ve bir prognostik faktör olmaya adaydır.

Aim: To determine whether mechanosensitive Piezo 1 and Piezo 2 channels are a potential marker in the diagnosis of endometrial cancer and ovarian cancer by determining the relative mRNA expressions in endometrial cancer and ovarian cancer. Materials and methods: Sections were taken from pathology blocks of ovarian and endometrial cancer patients who were operated on at Atatürk University Training and Research Hospital, and from pathology blocks containing healthy ovarian and endometrial tissue. Total RNA isolation was performed using a kit suitable for RNA extraction from the sections taken from the pathology blocks. Subsequently, cDNA synthesis was performed from the obtained RNAs. Piezo 1 and Piezo 2 gene expressions were determined by Real Time PCR (qPCR) method. Results: It was observed that both Piezo 1 and Piezo 2 showed a significant increase in expression in endometrial cancer tissue compared to normal endometrial tissue. There was a statistically significant correlation between tumor volume, lymphovascular invasion and p53 expression and Piezo 1 expressions according to the pearson correlation tests. It was observed that both Piezo 1 and Piezo 2 showed a significant increase in expression in ovarian cancer tissue compared to normal ovarian tissue. Although there was no statistically significant pathology feature according to Pearson correlation tests, preoperative Piezo 1 and Piezo 2 expressions showed weak-moderate and moderate correlations with some pathological values. Conclusion: It has been observed that Piezo 1 and Piezo 2 expressions are correlated with various clinical markers, clinical conditions and some pathological features, together with an increase in expression in endometrial cancer and ovarian cancer compared to healthy tissues. Piezo 1 and Piezo 2 are candidates for being a biomarker and a prognostic factor for endometrial cancer and ovarian cancer, although it is still very early.
