The use of fast, economical and environmentally friendly technologies to ensure food safety is important in terms of supporting sustainable agriculture. Escherichia coli is a pathogen that threat to food safety, prohibited in food products and must be monitored. For this purpose; counting methods or some molecular-based analyses can be applied. However, these methods are time-consuming, expensive, and require suitable laboratory conditions, experts in their fields and equipment. Also, they are not sustainable due to the need for excessive chemical use. Additionally, they can yield inaccurate results or encounter difficulties in detecting very low concentrations. Therefore, biosensor technologies, which are highly accurate, fast, user-friendly, and sensitive analytical devices, offer advantages for E. coli detection. In this study, a paper-based, cost- effective, flexible and disposable electrochemical biosensor for detecting E. coli was developed. A polyclonal E. coli antibody was preferred for the biosensor to be specific to E. coli; a multi-walled carbon nanotube (MWCNT) was used to increase the conductivity and the antibody and MWCNT were covalently bonded and applied as a conjugate. As a result of optimization studies, flexible electrodes were prepared using photographic paper printed with a 4B pencil tip and a cutting/printing device. As a result of the study, a biosensor with a detection range of 10 0 –10 7 CFU/mL and a LOD value of 26 CFU/mL was developed. Additionally, the biosensor successfully detected E. coli in various culture environments, including cultures of Bacillus subtilis, Saccharomyces cerevisiae, Lactiplantibacillus plantarum, and mixed cultures of all strains. Apple juice was used as the food matrix and successful results were also obtained in real sample trials. The biosensor was able to detect E. coli isolated directly from food samples, in addition to commercial cultures. This study is the first to develop a paper-based biosensor for E. coli detection using a pencil-drawing method and it is expected to lead future studies.
