Özet

Amaç: Onopordum acanthium L., yaygın adıyla“İskoç devedikeni,”Asteraceae familyasına ait iki yıllık bir bitki türüdür. Avrupa ve Asya'ya özgü olan bu bitki, zamanla Amerika ve Avustralya'ya da yayılmıştır. O. acanthium, anti-inflamatuar, antitümör ve kardiyotonik etkiler gibi dikkate değer tıbbi özellikleriyle tanınmaktadır. Bu çalışmanın temel amacı, bitkinin farmakolojik etkilerini inceleyerek hastalıkların önlenmesi ve tedavisindeki potansiyel rolünü araştırmaktır. Materyal ve Metot: Bitkinin 5 farklı kısmının (kök, gövde, çiçek, meyve ve yaprak) metanol ve su ekstreleri hazırlanmış, fenolik içerikleri LC-MS/MS kullanılarak belirlenmiştir. Ekstrelerin toplam fenolik madde miktarı için Folin-Ciocalteau yöntemi, antioksidan aktivite için DPPH• ve ABTS•+ süpürücü etki yöntemleri, antidiyabetik aktivite deneylerinde α-glukozidaz ve α-amilaz inhibisyon etki deneyleri, antikolinesteraz etki deneyleri için asetilkolinesteraz ve butirilkolinesteraz inhibisyon deneyleri, antibakteriyel ve antifungal aktivite tayinleri için mikrodilüsyon yöntemi kullanılmıştır. Numunelerdeki sekonder metabolitlerin kalitatif ve kantitatif analizleri, Atatürk Üniversitesi Doğu Anadolu Yüksek Teknoloji Uygulama ve Araştırma Merkezi (DAYTAM)'nde kromatografi ve spektroskopi sistemlerinin birleştirilmesiyle oluşturulan Agilent 6460 Triple Quadrupole Sıvı Kromatografi-Tandem Kütle/Kütle Spektrometre Sistemi (LC-MS/MS) kullanılarak gerçekleştirilmiştir. Bulgular: Bu çalışmada Onopordum acanthium bitkisinin çiçek, meyve, gövde, yaprak ve kök kısımları kullanılmıştır. Hazırlanan ekstrelerin miktarları sırasıyla şu şekilde ölçülmüştür: çiçek su ekstresi (5.3 mg), çiçek metanol ekstresi (5.6 mg), meyve su ekstresi (5.8 mg), meyve metanol ekstresi (5.6 mg), gövde su ekstresi (5.7 mg), gövde metanol ekstresi (5.8 mg), yaprak su ekstresi (6 mg), yaprak metanol ekstresi (5.2 mg), kök su ekstresi (5.6 mg) ve kök metanol ekstresi (5.5 mg). Bitki kısımlarında yapılan analizler sonucunda ana bileşenler şu şekilde tespit edilmiştir: kinik asit (804.6046 ng/mL), fumarik asit (46.1894 ng/mL), kerasiyanin klorür (406.4505 ng/mL), siyanidin-3-O-glikozit (222.2524 ng/mL), klorojenik asit (3045.3840 ng/mL), 4-hidroksibenzoik asit (2.6839 ng/mL), vanilik asit (46.7281 ng/mL), şirinjik asit (11.2655 ng/mL), ferulik asit (121.6395 ng/mL), rosmarinik asit (100.4060 ng/mL), vanilin (44.4214 ng/mL), resveratrol (3.0968 ng/mL) ve kersetin (1249.9104 ng/mL). Yapılan aktivite çalışmaları, O. acanthium bitkisinin farklı ekstrelerinin çeşitli hastalıklar üzerindeki etkilerinin farklılık gösterdiğini ortaya koymuştur. Sonuç: Onopordum acanthium L. bitkisinin farklı kısımlarından elde edilen metanol ve su ekstreleri, zengin fenolik içeriği ve belirgin biyolojik aktiviteleriyle dikkat çekmiştir. Elde edilen sonuçlar, bu bitkinin potansiyel antioksidan, antikolinesteraz, antidiyabetik ve antimikrobiyal etkiler sunduğunu göstermektedir.

Aim: Onopordum acanthium L., commonly known as“Scottish thistle,”is a biennial plant species belonging to the Asteraceae family. Native to Europe and Asia, this plant has gradually spread to America and Australia. O. acanthium is renowned for its notable medicinal properties, including anti-inflammatory, antitumor, and cardiotonic effects. The primary aim of this study is to investigate the pharmacological effects of the plant and explore its potential role in the prevention and treatment of diseases. Materials and Methods: Methanol and water extracts were prepared from five different parts of the plant (root, stem, flower, fruit, and leaf), and their phenolic contents were determined using LC-MS/MS. The Folin-Ciocalteu method was employed to measure the total phenolic content of the extracts, while antioxidant activities were evaluated using DPPH• and ABTS•+ radical scavenging assays. Antidiabetic activity was assessed through α-glucosidase and α-amylase inhibition assays, and anticholinesterase activity was examined via acetylcholinesterase and butyrylcholinesterase inhibition experiments. Antibacterial and antifungal activities were determined using the microdilution method. Qualitative and quantitative analyses of secondary metabolites in the samples were performed at the Eastern Anatolia High Technology Application and Research Center (DAYTAM) of Atatürk University, utilizing the Agilent 6460 Triple Quadrupole Liquid Chromatography-Tandem Mass Spectrometry System (LC-MS/MS), which integrates chromatography and spectroscopy systems. Results: In this study, the flower, fruit, stem, leaf, and root parts of Onopordum acanthium were utilized. The amounts of the prepared extracts were measured as follows: flower water extract (5.3 mg), flower methanol extract (5.6 mg), fruit water extract (5.8 mg), fruit methanol extract (5.6 mg), stem water extract (5.7 mg), stem methanol extract (5.8 mg), leaf water extract (6 mg), leaf methanol extract (5.2 mg), root water extract (5.6 mg), and root methanol extract (5.5 mg). The main components identified through analyses of the plant parts were as follows: quinic acid (804.6046 ng/mL), fumaric acid (46.1894 ng/mL), keracyanin chloride (406.4505 ng/mL), cyanidin-3-O-glucoside (222.2524 ng/mL), chlorogenic acid (3045.3840 ng/mL), 4-hydroxybenzoic acid (2.6839 ng/mL), vanillic acid (46.7281 ng/mL), syringic acid (11.2655 ng/mL), ferulic acid (121.6395 ng/mL), rosmarinic acid (100.4060 ng/mL), vanillin (44.4214 ng/mL), resveratrol (3.0968 ng/mL), and quercetin (1249.9104 ng/mL). The activity studies revealed that the effects of different extracts of O. acanthium on various diseases varied significantly. Conclusion: The methanol and water extracts obtained from different parts of Onopordum acanthium L. have shown rich phenolic content and remarkable biological activities. The results indicate that this plant offers potential antioxidant, anticholinesterase, antidiabetic, and antimicrobial effects.
