Özet
Giriş ve Amaç: Multiple skleroz (MS) dünya çapında 2,8 milyondan fazla kişiyi etkilemekte olup karmaşık gen çevre etkileşimi nedeniyle etiyolojisi tam olarak bilinmemektedir. MS'e katkıda bulunduğu düşünülen çok sayıda gen grubu bulunmaktadır. MS'in patogenezinde santral sinir sisteminde önemli rolleri olan folat ve homosistein mekanizmasının etkili olduğu düşünülmektedir. MS riski olan kişilerde hastalığın erken tanısı ve tedavisine katkı sağlayabilecek genetik etkilerin belirlenmesi için folat ve homosistein mekanizmalarını etkileyebileceğini düşündüğümüz MTR (A2756G), MTRR (A66G) ve MTHFR (C677T) gen varyasyonlarını araştırmayı amaçladık. Gereç ve Yöntemler: Bu çalışmada 102 MS tanılı hastadan ve 102 sağlıklı kontrol grubundan alınan venöz kandan DNA örnekleri elde edildi. Elde edilen DNA örneklerinden polimeraz zincir reaksiyonu (PZR) ve retriksiyon fragment uzunluk polimorfizmi (RFUP) yöntemleri kullanılarak MTR (A2756G), MTRR (A66G), MTHFR (C677T) gen varyasyonları genotip dağılımları bulunarak hastalığa yatkınlık belirlendi. Bulgular: Multiple skleroz hastalarının ailesinde MS görülme oranı sağlıklı kontrol grubuna oranla istatistiksel açıdan anlamlı olarak yüksek bulundu. MS'li hasta ve sağlıklı kontrol grupları arasında MTR (A2756G) ve MTHFR (C677T) gen varyasyonlarının genotip dağılımında anlamlı bir fark bulunamadı. MTRR (A66G) genotip dağılımında MS tanılı grupta AA genotipi kontrol grubuna oranla istatistiksel açıdan anlamlı olarak yüksek bulundu. MTRR (A66G) AG genotipi ise kontrol grubunda hasta grubuna oranla istatistiksel açıdan anlamlı olarak yüksek bulundu. Sonuç: Çalışmamız sonuçlarına göre ailesinde multiple skleroz olan kişilerin MS riskinin arttığı, MTRR (A66G) gen varyasyonunun AA genotipinin MS hastalığına karşı risk oluşturabileceği ve MTRR (A66G) AG genotipinin MS karşı koruyucu faktör olabileceğini düşünmekteyiz.

Background and Aim: Multiple sclerosis (MS) affects more than 2.8 million people worldwide and its etiology is not fully known due to complex gene-environment interactions. There are many gene groups thought to contribute to MS. It is thought that folate and homocysteine mechanisms, which play important roles in the central nervous system, are effective in the pathogenesis of MS. We aimed to investigate MTR (A2756G), MTRR (A66G) and MTHFR (C677T) gene variations, which we think may affect folate and homocysteine mechanisms, in order to determine genetic effects that may contribute to the early diagnosis and treatment of the disease in people at risk for MS. Materials and Methods: In this study, DNA samples were obtained from venous blood taken from 102 patients diagnosed with MS and 102 healthy controls. From the obtained DNA samples, genotype distributions of MTR (A2756G), MTRR (A66G), MTHFR (C677) gene variations were found using polymerase chain reaction (PCR) and retraction fragment length polymorphism methods and predisposition to the disease was determined. Material and Methods: In this study, DNA samples were obtained from venous blood taken from 102 patients diagnosed with MS and 102 healthy controls. The genotype distributions of MTR, MTRR, MTHFR gene variations were found using polymerase chain reaction (PCR) and retraction fragment length polymorphism (RFLP) methods from the obtained DNA samples and the predisposition to the disease was determined. Results: The incidence of MS in the families of multiple sclerosis patients was found to be statistically significantly higher than in the healthy control group. No significant difference was found in the genotype distribution of MTR (A2756G) and MTHFR (C677T) gene variations between the MS patient and healthy control groups. In the MTRR (A66G) genotype distribution, the AA genotype was found to be statistically significantly higher in the MS diagnosed group than in the control group. The MTRR (A66G) AG genotype was found to be statistically significantly higher in the control group than in the patient group. Conclusion: According to the results of our study, we think that people with multiple sclerosis in their family have an increased risk of MS, that the AA genotype of the MTRR (A66G) gene variation may pose a risk for MS disease, and that the MTRR (A66G) AG genotype may be a protective factor against MS.
