Özet
Bu çalışmanın amacı, kritik altı su ekstraksiyonu (KASE) yöntemiyle Red Delicious cinsi elmaların kabuklarından fenolik bileşiklerin özütlenmesi ve ekstraksiyon sıcaklığı, etanol oranı ve süre gibi koşulların, özütlerin toplam fenolik madde (TFMİ), flavonoid içeriği (TFİ) ve antioksidan aktiviteleri (AA) üzerine etkisinin belirlenmesidir. Araştırmada hem laboratuvar ölçekli KASE sistemi hem de hızlandırılmış solvent ekstraksiyonu (ASE) yöntemi kullanılmıştır. ASE yönteminin merkezi karma tasarım deseniyle optimize edildiği çalışmada, her üç yanıt faktörünün maksimize edildiği optimum koşul yaklaşık 108,5°C, %2,03 çözücü etanol oranı ve 26 dakikalık süre olmuştur. Bu koşullarda elde edilen ekstraktların TFİ, TFMİ ve AA değerleri sırasıyla 45,01 mg KE/L, 77,67 mg GAE/L ve 52,49 µmol TE/L olarak belirlenmiştir. Optimum koşullardaki ekstraktın biyaktif özellikleri geleneksel ve ultrasonikasyon destekli ekstraksiyon (UDE) yöntemleriyle elde edilenlerle kıyaslanmıştır. ASE ve geleneksel yöntemle elde edilen ekstraktların TFMİ değerleri sırasıyla 13,82 ve 14,22 mg GAE/g olurken (p>0,05), UDE yöntemi en düşük TFMİ değerini (1,80 mg GAE/g) vermiştir (p<0,05). ASE (8,95 µmol TE/g) ve UDE (8,72 µmol TE/g) yöntemleriyle elde edilen ekstraktların AA değerleri benzer bulunurken (p>0,05), geleneksel ekstraksiyonda daha düşük AA değeri (4,64 µmol TE/g) belirlenmiştir. Ekstraktların TFİ açısından ise UDE 10,68 mg KE/g ile en yüksek değeri verirken (p<0,05), ASE (6,58 mg KE/g) ve geleneksel ekstraksiyon (5,64 mg KE/g) yöntemleri daha düşük değerler göstermiştir. Çalışma bulguları, KASE yönteminin, fenolik bileşiklerin verimli bir şekilde özütlenmesinde etkili bir yöntem olarak öne çıktığını ve optimize edilen koşullarda elde edilen ekstraktların biyolojik aktivitelerinin diğer ekstraksiyon yöntemleriyle kıyaslandığında üstün veya benzer performans sergilediğini göstermiştir.

The aim of this study is to extract phenolic compounds from the peels of Red Delicious apples using the subcritical water extraction (SWE) method and to determine the effects of conditions such as extraction temperature, ethanol ratio, and duration on the total phenolic content (TPC), flavonoid content (TFC), and antioxidant activities (AA) of the extracts. Both a laboratory-scale SWE system and the accelerated solvent extraction (ASE) method were utilized in the research. In the study, where the ASE method was optimized using a central composite design, the optimal conditions, under which all three response factors were maximized, were determined to be approximately 108.5°C, 2.03% solvent ethanol ratio, and a duration of 26 minutes. Under these conditions, the TFC, TPC, and AA values of the extracts were determined as 45.01 mg CE/L, 77.67 mg GAE/L, and 52.49 µmol TE/L, respectively. The bioactive properties of the extract obtained under optimal conditions were compared with those obtained using conventional and ultrasound-assisted extraction (UAE) methods. The TPC values of the extracts obtained by ASE and the conventional method were 13.82 and 14.22 mg GAE/g, respectively (p>0.05), while the UAE method yielded the lowest TPC value (1.80 mg GAE/g) (p<0.05). The AA values of the extracts obtained by ASE (8.95 µmol TE/g) and UAE (8.72 µmol TE/g) methods were found to be similar (p>0.05), whereas the conventional extraction method resulted in a lower AA value (4.64 µmol TE/g). In terms of TFC, UAE provided the highest value of 10.68 mg CE/g (p<0.05), while ASE (6.58 mg CE/g) and conventional extraction (5.64 mg CE/g) methods showed lower values. The findings of the study highlight that the SWE method is an effective approach for efficiently extracting phenolic compounds and that the extracts obtained under optimized conditions exhibit superior or comparable biological activities when compared to other extraction methods.
