Özet
Bu çalışmanın amacı, Mitomisin C (MMC) genotoksisitesine karşı Ribes rubrum L. (Rr, kırmızı frenk üzümü) meyve ve yaprak ekstraktının olası genotoksik ve antigenotoksik etkilerini in vivo olarak araştırmaktır. Bu kapsamda kırmızı frenk üzümüne ait meyve ve yaprak örnekleri, Temmuz-Ağustos 2024 tarihleri arasında Kars'ın Kağızman ilçesinden toplandı. Toplanan örneklerden, Soxhlet ekstraksiyon yöntemi kullanılarak etanolik meyve ve yaprak ekstraktları hazırlandı. Kırmızı frenk üzümü meyve ve yaprak örneklerine ait uçucu yağ bileşenleri GC-MS, fenolik bileşik içeriği ise LC-HRMS yöntemi ile analiz edildi. Meyve ve yaprak ekstraktının olası antigenotoksik-genotoksik potansiyeli, Sprague Dawley ırkı erkek rat kemik iliği kullanılarak Mikronükleus (MN) ve Kromozomal Aberasyon (KA) test yöntemleri ile belirlendi. Bu çalışma için 10 grup oluşturuldu; 1. grup: negatif kontrol grubu (NK), 2. grup: MMC grubu (2 mg/kg), 3. grup: 100 mg/kg Rr yaprak grubu, 4. grup: 100 mg/kg Rr meyve grubu, 5. grup: 500 mg/kg Rr yaprak grubu, 6. grup: 500 mg/kg Rr meyve grubu, 7. grup:100 mg/kg Rr yaprak+MMC grubu, 8. grup:100 mg/kg Rr meyve+MMC grubu, 9. grup: 500 mg/kg Rr yaprak+MMC, 10. grup: 500 mg/kg Rr meyve+MMC grubu. Kırmızı frenk üzümü meyve ve yaprak ekstraktı 15 gün boyunca oral yolla verildi. MMC uygulaması servikal dislokasyondan 24 saat öncesinde rat gruplarına intraperitonal (ip) olarak 2 mg/kg olarak verildi. Uygulama işleminden sonra, disloke edilen ratların femur kemik iliği hücrelerinde kromozomal aberasyon, mitotik indeks ve mikronükleus oranları tespit edildi. GC-MS analizinde, yaprak uçucu yağında toplam 31 bileşen (% 70,5 toplam uçucu bileşik oranı) ve meyve uçucu yağında ise toplam 21 bileşen (% 81,8 toplam uçucu bileşik oranı) tespit edildi. Bu bileşenler arasında yaprakta yüksek oranda heptokosan (% 6,7) ve meyvede ise hekzadekanoik asit (palmitik asit) (% 44,1) olduğu saptandı. LC-HRMS yöntemi ile yapılan fenolik bileşik analizinde; yaprak ekstraktında 32, meyve ekstraktında ise 13 fenolik bileşiğin varlığı tespit edildi. Yaprak ve meyve ekstraktında yüksek oranda bulunan bileşikler sırasıyla nikotiflorin (9.280,40 ng/mL) ve rutin hidrat M-OH2 (341,20 ng/mL) olarak belirlendi. MMC uygulaması, kromozomal aberasyon ve mikronükleus oranlarında istatistiksel olarak anlamlı artışlara (p<0.001) ve mitotik indeks oranında ise anlamlı azalmaya (p<0.001) yol açmıştır. Bu durum MMC'nin genotoksik etkili bir ajan olduğunu açıkça ortaya koymaktadır. Tek başına uygulanan 100 mg/kg ve 500 mg/kg dozlarındaki Rr meyve ve yaprak ekstraktları ise negatif kontrol grubuna benzer sonuçlar göstererek herhangi bir genotoksik etki oluşturmamıştır (p>0.05). MMC ile birlikte uygulanan meyve ve yaprak ekstraktları, özellikle 500 mg/kg yaprak ekstraktı dozunda, MMC'nin neden olduğu kromozomal aberasyon ve mikronükleus oranlarını istatistiksel olarak anlamlı düzeyde azaltmış (p<0.001) ve mitotik indeks oranını artırmıştır (p<0.001). Sonuç olarak, fenolik bileşikler ve uçucu yağ içeriği bakımından zengin olan kırmızı frenk üzümü meyve ve yaprak ekstraktları, kemoterapötik bir ajan olan MMC'nin oluşturduğu genotoksisiteyi azaltmada potansiyel bir doğal koruyucu ajan olarak değerlendirilebilir.

The present study aimed to investigate the potential genotoxic and antigenotoxic effects of Ribes rubrum L. (Rr, red currant) fruit and leaf extracts in vivo against genotoxicity induced by Mitomycin C (MMC). For this purpose, red currant fruit and leaf samples were collected from Kağızman, Kars, during the period of July–August 2024. Ethanol extracts of the fruit and leaf samples were prepared separately using the soxhlet extraction method. The volatile oil components of the red currant samples were analyzed using GC-MS, while the phenolic compound profiles were determined through LC-HRMS. To assess the genotoxic and antigenotoxic potential of the extracts, the Micronucleus (MN) and Chromosomal Aberration (CA) tests were applied using bone marrow cells obtained from male Sprague Dawley rats. Ten experimental groups were designed as follows: Group 1: negative control (NC), Group 2: MMC-treated (2 mg/kg, intraperitoneally), Group 3: Rr leaf extract (100 mg/kg), Group 4: Rr fruit extract (100 mg/kg), Group 5: Rr leaf extract (500 mg/kg), Group 6: Rr fruit extract (500 mg/kg), Group 7: Rr leaf extract (100 mg/kg) + MMC, Group 8: Rr fruit extract (100 mg/kg) + MMC, Group 9: Rr leaf extract (500 mg/kg) + MMC, and Group 10: Rr fruit extract (500 mg/kg) + MMC. The extracts were administered orally for 15 consecutive days, and MMC was injected intraperitoneally at a dose of 2 mg/kg, 24 hours prior to cervical dislocation. Following the treatments, chromosomal aberrations, mitotic index, and micronucleus frequencies were evaluated in femoral bone marrow cells. According to the GC-MS results, a total of 31 compounds were identified in the leaf essential oil, accounting for 70.5% of the total volatile content, while 21 compounds were detected in the fruit oil, representing 81.8% of the total volatile content. The major components were heptacosane (6.7%) in the leaf oil and hexadecanoic acid (palmitic acid, 44.1%) in the fruit oil. LC-HRMS analysis revealed the presence of 32 phenolic compounds in the leaf extract and 13 in the fruit extract. Among these, nicotiflorin (9,280.40 ng/mL) and rutin hydrate M-OH₂ (341.20 ng/mL) were the most abundant phenolic constituents in the leaf and fruit extracts, respectively. MMC treatment significantly increased the frequency of CA and MNPCE (p<0.001), and caused a significant decrease in the mitotic index (p<0.001), confirming its genotoxic effects. In contrast, administration of red currant fruit and leaf extracts alone at both 100 mg/kg and 500 mg/kg doses did not result in any statistically significant genotoxicity compared to the negative control group (p>0.05). Notably, co-treatment with red currant extracts and MMC particularly the 500 mg/kg dose of the leaf extract led to a statistically significant reduction in MMC induced chromosomal aberrations and micronucleus formation (p<0.001), as well as a significant increase in mitotic index (p<0.001). In conclusion, red currant fruit and leaf extracts, which are rich in phenolic compounds and volatile constituents, demonstrated a protective effect against MMC induced genotoxicity in rats. These findings suggest that Ribes rubrum may be considered a promising natural agent for mitigating genotoxic damage caused by chemotherapeutic agents.
