Özet
Kolon kanseri, dünyada en sık görülen üçüncü kanser türüdür. Hem tekrarlama riskinin yüksek olması hem de hastalığın heterojen yapısı nedeniyle, kolon kanseri tanı ve tedavisinde yeni moleküler belirteçlerin keşfi önem taşımaktadır. Spalt benzeri (SALL) gen ailesi, transkripsiyon faktörleri olarak işlev görür ve ürettikleri proteinlerle hücre proliferasyonu, farklılaşma ve kök hücre benzeri özelliklerin kontrolü de dahil olmak üzere gen ekspresyonunun düzenlenmesinde rol alırlar. Güncel çalışmalarda mikroRNA'ların, gen regülasyonunda önemli görevleri olduğu ve karsinogenez sürecine katkı sağladığı bilinmektedir. Çalışmada, 25 kolon kanser hastasına ait tümör dokusu ve komşu normal doku örneklerinde SALL1, SALL2, SALL3 ve SALL4 genleri ile miR-5688, miR-1305, miR-130a-5p ve miR-1226-3p ekspresyon düzeyleri dijital Polimeraz Zincir Reaksiyonu yöntemiyle analiz edilmiştir. Tümör dokusu ve komşu normal doku arasındaki SALL2 ve SALL3 genlerinin anlamlı ekspresyon farkları (p<0,001; p=0,008) ve yüksek ROC değerleri (AUC=0,818; 0,718), bu genlerin biyobelirteç adayı olma potansiyelini güçlendirmektedir. SALL2 ve SALL3 genlerinin kombine kullanımı, kolon kanserinde histopatolojik tanıyı destekleyecek biyobelirteç panelleri için potansiyel olarak temel oluşturabilir. Ayrıca SALL1 ve SALL2 gen ekspresyonları arasında saptanan yüksek pozitif korelasyonda (r=0,601; p=0,002), bu genlerin kombine biyobelirteç panelinde birlikte değerlendirilme potansiyelini desteklemektedir. SALL4 geni, miR-1226-3p ve miR-1305 ekspresyon düzeylerinin çoğu kolon dokusunda tespit edilememesi, ekspresyonlarının epigenetik baskılanma olasılığına işaret etmektedir. Bu bulgular, söz konusu moleküllerin kolon kanserinde sınırlı ekspresyon profiline sahip olabileceğini göstermektedir. Çalışma sonuçları, SALL2 ve SALL3 genlerinin kolon kanserinde tanısal biyobelirteç olma potansiyeline işaret eden ön veriler sunmaktadır. Kesin sonuçlara varılabilmesi için bu bulguların daha geniş ve çok merkezli gruplarda doğrulanması gerekmektedir.

Colon cancer is the third most commonly diagnosed cancer type worldwide. Due to its high recurrence rate and heterogeneous nature, the identification of novel molecular biomarkers is crucial for the diagnosis and treatment of colon cancer. The Spalt-like (SALL) gene family functions as transcription factors and plays a role in the regulation of gene expression through the proteins they encode, particularly in processes such as cell proliferation, differentiation, and the maintenance of stem cell-like properties. Recent studies have shown that microRNAs play important roles in gene regulation and contribute to the process of carcinogenesis. In this study, expression levels of SALL1, SALL2, SALL3, and SALL4 genes, as well as miR-5688, miR-1305, miR-130a-5p, and miR-1226-3p, were analyzed using digital Polymerase Chain Reaction in tumor and adjacent normal tissue samples obtained from 25 colon cancer patients. Significant expression differences of SALL2 and SALL3 genes between tumor and adjacent normal tissues (p<0,001; p=0,008), along with high ROC values (AU =0,818; 0,718), support the potential of these genes as biomarker candidates. The combined use of SALL2 and SALL3 genes may form a basis for biomarker panels that support histopathological diagnosis in colon cancer. Additionally, the strong positive correlation observed between SALL1 and SALL2 gene expressions (r=0,601; p=0,002) further supports their potential for joint evaluation in such panels. The lack of detectable expression of the SALL4 gene, miR-1226-3p, and miR-1305 in most colon tissue samples suggests possible epigenetic silencing of these molecules. These findings indicate that the mentioned molecules may exhibit a limited expression profile in colon cancer. The results of this study provide preliminary evidence supporting the diagnostic biomarker potential of SALL2 and SALL3 genes in colon cancer. However, validation of these findings in larger, multicenter cohorts is necessary to draw definitive conclusions.
