Özet

Kanser, günümüzde tüm dünyada artan görülme sıklığı ve yüksek mortalite oranlarıyla insan sağlığını tehdit eden en önemli hastalıklardan biri olarak öne çıkmaktadır. Konvansiyonel tedavi yöntemlerinin (kemoterapi, radyoterapi vb.) çeşitli yan etkiler barındırması, tedaviye karşı direnç gelişmesi ve hedef dışı hücrelerin zarar görmesi gibi nedenlerle daha etkili, güvenli ve hedefe yönelik alternatif terapilere olan ihtiyaç giderek artmaktadır. Bu doğrultuda fotodinamik terapi (PDT), sonodinamik terapi (SDT) ve bu iki yöntemin sinerjik bir kombinasyonu olan sono-fotodinamik terapi (SPDT), dikkat çeken yenilikçi yaklaşımlar olarak değerlendirilmektedir. PDT, uygun bir fotosensitizerin ışık ile uyarılması sonucu singlet oksijen gibi reaktif oksijen türlerinin (ROS) oluşturulmasına dayanan bir tedavi yöntemi iken; SDT'de benzer bir etki ses dalgaları (ultrason) aracılığıyla sağlanmaktadır. SPDT yöntemi ise her iki tedavi metodunun avantajlarını birleştirerek daha güçlü ve etkili bir terapötik sonuç elde etmeyi hedeflemektedir. SPDT yönteminin başarısı, doğrudan kullanılan sono-fotosensitizer bileşiğin özelliklerine bağlıdır. Bu nedenle, antikanser uygulamalarda yalnızca tedavi yöntemi değil, aynı zamanda seçilen sensitizerin yapısal ve fotokimyasal özellikleri de büyük önem taşımaktadır. Bu bağlamda ftalosiyanin türevleri; görünür bölgedeki (600–800 nm) yüksek molar absorbsiyon katsayıları, uzun triplet durum ömürleri, geniş konjuge π-elektron sistemleri, fotokimyasal kararlılıkları, toksik olmamaları ve iyi çözünürlük özellikleri sayesinde öne çıkan adaylardır. Geçiş metalleri içeren metallo-ftalosiyanin kompleksleri, uygun sübstitüentlerle modifiye edildiğinde hem hücre hedeflenebilirliği hem de yüksek singlet oksijen üretimi açısından umut verici bileşikler olarak değerlendirilmektedir. Bu çalışmada, 4-klorotiyofenol ve 4-bromotiyofenol gruplarıyla sübstitüe edilmiş çinko(II) ftalosiyanin kompleksleri tasarlanmış, sentezlenmiş ve UV/Vis, FT-IR, ¹H-NMR ve MALDI-TOF-MS teknikleriyle detaylı yapısal karakterizasyonları gerçekleştirilmiştir. Yürütülen bu çalışma ile yeni nesil sono-fotosensitizerlerin tasarlanarak sentezlenmesi ve bu bileşiklerin fotokimyasal ve sono-fotokimyasal özelliklerinin değerlendirilmesi hedeflenmiştir. Elde edilen sonuçlar, geliştirilen ftalosiyanin türevlerinin yüksek verimli singlet oksijen üretme potansiyeline sahip olduğunu ve SPDT uygulamaları için uygun adaylar olduğunu göstermektedir. Bu bileşiklerin ileri düzey biyolojik testlerde (in vitro/in vivo) değerlendirilmesi, kanser tedavisine yönelik yeni nesil, hedeflenebilir ve daha az yan etkiye sahip tedavi seçeneklerinin geliştirilmesine katkı sağlayacaktır. Ayrıca bu çalışma; kimya, biyoloji, farmakoloji ve tıp gibi disiplinlerarası alanlarda yürütülecek çok sayıda yeni araştırmaya da zemin oluşturabilecek niteliktedir.

Cancer remains one of the most serious global health challenges of our time, characterized by its increasing incidence and high mortality rates. Due to the various side effects of conventional treatment methods (such as chemotherapy and radiotherapy), the development of resistance, and the damage caused to non-target tissues, there is a growing need for more effective, safer, and targeted alternative therapies. In this context, photodynamic therapy (PDT), sonodynamic therapy (SDT), and the synergistic combination of these two methods—sonophotodynamic therapy (SPDT)—have emerged as some of the most promising innovative approaches. PDT involves the excitation of a suitable photosensitizer by light to generate reactive oxygen species (ROS), such as singlet oxygen, whereas SDT achieves similar effects through the application of sound waves (ultrasound). SPDT aims to combine the advantages of both methods, thereby achieving more potent and effective therapeutic outcomes. The success of SPDT depends largely on the properties of the sono-photosensitizer employed. Therefore, in anticancer applications, not only the therapeutic approach but also the structural and photochemical characteristics of the sensitizer are of critical importance. In this regard, phthalocyanine derivatives stand out as promising candidates due to their strong absorption in the visible region (600–800 nm), long triplet-state lifetimes, extensive conjugated π-electron systems, photochemical stability, non-toxicity, and favorable solubility. Metallo-phthalocyanine complexes containing transition metals are considered particularly promising compounds in terms of both cell-targeting ability and high singlet oxygen generation when appropriately substituted. In this study, zinc(II) phthalocyanine complexes substituted with 4-chlorothiophenol and 4-bromothiophenol groups were designed, synthesized, and thoroughly characterized using UV/Vis, FT-IR, ¹H-NMR, and MALDI-TOF-MS techniques. This work aimed to design and synthesize new-generation sono-photosensitizers and to evaluate their photochemical and sonophotochemical properties. The results demonstrated that the developed phthalocyanine derivatives possess high singlet oxygen generation efficiency, indicating their potential suitability for SPDT applications. Further biological evaluations (in vitro and in vivo) of these compounds could contribute to the development of next-generation, targeted cancer therapies with fewer side effects. Moreover, this study provides a valuable foundation for future interdisciplinary research in fields such as chemistry, biology, pharmacology, and medicine.
