Özet
Bu tez çalışmasında, bal arısı (Apis mellifera) kolonilerinde ekonomik kayıplara neden olan ektoparazitik akar Varroa destructor'a karşı biyolojik mücadelede kullanılabilecek yerel mikrobiyal izolatların izolasyonu ve karakterizasyonu amaçlanmıştır. Araştırmada, V. destructor bireyleri İzmir Menemen'den, parazitli ve sağlıklı A. mellifera bireyleri ise İzmir Kemalpaşa'daki bir arı çiftliğinden temin edilmiştir. Çalışma, laboratuvar ölçeğinde yürütülmüş ve deneysel tasarım doğrultusunda üç ana aşamadan oluşmuştur: mikrobiyal izolasyon ve fenotipik karakterizasyon, biyotest uygulamaları ve moleküler tanımlama. İlk aşamada akar bireylerinden yedi farklı mikrobiyal izolat elde edilmiş ve fenotipik özellikleri temel testlerle değerlendirilmiştir. Bu izolatların olası biyolojik etkilerini belirlemek amacıyla, ilk olarak hareketlilik biyotesti uygulanmıştır. Her bir izolatın V. destructor bireyleri üzerindeki etkisi 0., 1., 2., 3., 4. ve 5. saatlerde gözlemlenmiş; bu test sonucunda M6KA ve M6AB kodlu izolatlar yüksek düzeyde etkili bulunmuştur. İkinci aşamada, seçilen bu iki izolatın ayrı ayrı ve konsorsiyum halinde, arı bireyleri üzerindeki etkileri 7 gün boyunca sürdürülen kafes denemeleri ile test edilmiştir. Her bir kafeste 20 arı bulunan altı deney grubu oluşturulmuş; bu grupların üçü varroalı, üçü sağlıklı arılardan seçilmiştir. Denemeler, McFarland 1.0 standardına uygun bakteri süspansiyonlarının püskürtme yöntemiyle uygulanması şeklinde gerçekleştirilmiştir. Elde edilen bulgulara göre, hem M6KA hem de M6AB izolatları varroalı arılar üzerinde belirgin mortalite artışına neden olmuştur. Bununla birlikte, M6KA ile muamele edilen sağlıklı arılarda kontrol ve FTS gruplarına kıyasla daha yüksek mortalite oranları gözlenmiş, bu durum izolatın arı bireyleri açısından toksik bir profil sergileyebileceğini düşündürmüştür. M6AB izolatı ise varroalı arılarda etkili olurken, sağlıklı arılar üzerinde görece daha düşük mortalite ve bazı zaman noktalarında koruyucu etki ile dikkat çekmiştir. Her iki izolatın birlikte kullanıldığı konsorsiyum grubunda ise hem akar düşürücü etkinin korunduğu hem de sağlıklı arılarda yüksek sağ kalım oranlarının büyük ölçüde devam ettiği belirlenmiştir. Son aşamada, M6KA ve M6AB izolatları moleküler düzeyde tanımlanmıştır. 16S rRNA gen bölgesinin amplifikasyonu ve BLAST analizi sonucunda her iki izolatın Bacillus velezensis, Bacillus amyloliquefaciens ve M6KA için ayrıca Bacillus siamensis türleriyle yüksek benzerlik gösterdiği tespit edilmiştir. Bu türlerin filogenetik yakınlığı nedeniyle tür düzeyinde kesin ayrım yapmak zor olsa da, izolatların biyolojik mücadele potansiyelini destekler nitelikte olduğu görülmüştür. Bu çalışma, Türkiye'deki yerel mikrobiyal suşların V. destructor ile mücadelede çevre dostu ve alternatif biyolojik ajanlar olarak değerlendirilebileceğini ortaya koyarak literatüre katkı sağlamaktadır.

In this thesis study, it was aimed to isolate and characterize local microbial isolates that could be used in the biological control of the ectoparasitic mite V. destructor, which causes significant economic losses in Apis mellifera colonies. In the study, V. destructor specimens were obtained from Menemen, İzmir, and parasitized and healthy A. mellifera individuals were obtained from a beekeeping farm in Kemalpaşa, İzmir. The study was conducted on a laboratory scale and consisted of three main stages: microbial isolation and phenotypic characterization, bioassay applications, and molecular identification. In the first stage, seven different microbial isolates were obtained from mite individuals and their phenotypic characteristics were evaluated using basic tests. To determine the possible biological effects of these isolates, a mobility bioassay was initially applied. The effects of each isolate on V. destructor individuals were observed at the 0th, 1st, 2nd, 3rd, 4th, and 5th hours; as a result of this test, isolates coded M6KA and M6AB were found to be highly effective. In the second stage, the effects of these two selected isolates, individually and in consortium, on bee individuals were tested through cage trials conducted over 7 days. Six experimental groups, each consisting of 20 bees, were formed; three of these groups consisted of Varroa-infested bees, and the other three of healthy bees. The trials were conducted by applying bacterial suspensions, adjusted according to the McFarland 1.0 standard, via spraying method. According to the findings obtained, both M6KA and M6AB isolates caused a notable increase in mortality in Varroa-infested bees. However, healthy bees treated with M6KA exhibited higher mortality rates compared to the control and FTS groups, suggesting that this isolate may present a toxic profile for honey bee individuals. In contrast, while the M6AB isolate was effective against Varroa-infested bees, it showed relatively lower mortality in healthy bees and even demonstrated a protective effect at certain time points. In the consortium group, where both isolates were applied together, the Varroa-reducing effect was maintained, and high survival rates in healthy bees were largely preserved. In the final stage, M6KA and M6AB isolates were identified at the molecular level. As a result of amplification of the 16S rRNA gene region and BLAST analysis, both isolates were found to show high similarity with B. velezensis, B. amyloliquefaciens, and additionally for M6KA, B. siamensis. Although definitive species-level identification was difficult due to the phylogenetic closeness of these species, the findings support the biocontrol potential of the isolates. This study contributes to the literature by revealing that local microbial isolates from Türkiye may be evaluated as environmentally friendly and alternative biological control components against V. destructor.
