On dördüncü günde haşhaş yağı grubunda CK-16 ekspresyonunun fitokrem grubuna göre anlamlı düzeyde yüksek olması (P=0,01), geç dönemde epitel stres yanıtının sürdüğünü gösterdi. Yedinci günde kontrol grubuna kıyasla miR-21 ekspresyonunun haşhaş yağı grubunda yaklaşık 2.43 kat arttığı, 14. günde ise haşhaş yağı grubunda miR-21 ekspresyonunun kontrol grubuna göre yaklaşık 29 kat arttığı görüldü. Sonuç olarak, haşhaş yağının yara iyileşme süreci üzerindeki etkisi histopatolojik ve moleküler düzeyde değerlendirilmiş olup, elde edilen verilerin mevcut literatüre katkı sağlayarak gelecekte yapılacak çalışmalara yol gösterici nitelikte olacağı düşünülmektedir.
Özet (Çeviri)
In this study, a total of 40 female CD-1 mice, aged 4-5 weeks and weighing approximately 80 g on average, were used. The animals were randomly assigned into four groups: poppy seed oil, phytocream, antagomir21, and control. All groups were maintained under a 12-hour light/12-hour dark cycle, with ad libitum access to food and water. In all animals, a full-thickness excisional skin wound was created on the dorsal region using a 0,6 mm biopsy punch, positioned 0,5 cm to the right of the dorsal midline. Each experimental group was further divided into two subgroups corresponding to the 7th and 14th days. Throughout the study, the poppy seed oil group received topical application of poppy seed oil once daily at the same time each day. Similarly, the phytocream group was treated with topical phytocream once daily at the same time. In the antagomir21 group, antagomir-21 was injected into the periwound area at four different points immediately after wound induction, and poppy seed oil was additionally applied throughout the study period. No treatment was administered to the control group. Animals were euthanized on days 7 and 14, and skin samples were collected from the wound area. The obtained tissue specimens were evaluated using histopathological, immunohistochemical, and real-time polymerase chain reaction (RT-PCR) analyses. Macroscopic evaluation on day 7 demonstrated a statistically significant difference in wound area width between the poppy seed oil group and the antagomir21 group (P=0.045). For histopathological assessment, hematoxylin–eosin staining was used to evaluate re-epithelialization, epidermal thickness, granulation tissue formation, inflammation, angiogenesis, edema, and hemorrhage. Masson's trichrome staining was performed to assess collagen density, collagen organization, and scar tissue maturation. Based on the total scores obtained from these parameters, the group with the highest score was considered the most favorable in terms of wound healing. Histological scoring on day 7 revealed a statistically significant difference between the poppy seed oil and antagomir21 groups (P=0.018). Immunohistochemical analyses were performed using antibodies against VEGF, FGF, PDGF, CK-16, Ki-67, and CD-31. On day 7, VEGF expression in the poppy seed oil group was significantly higher than that observed in the antagomir21 group (P=0.013). In addition, a statistically significant difference in PDGF expression was detected between the poppy seed oil and antagomir21 groups (P=0.039). On day 14, CK-16 expression was significantly higher in the poppy seed oil group compared to the phytocream group (P=0,01), indicating the persistence of epithelial stress response during the late phase of wound healing. Molecular analyses revealed that miR-21 expression was approximately 2.43-fold higher in the poppy seed oil group than in the control group on day 7, whereas on day 14, miR-21 expression in the poppy seed oil group increased by approximately 29-fold compared to the control group. In conclusion, the effects of poppy seed oil on the wound healing process were evaluated at histopathological, immunohistochemical, and molecular levels. The findings of this study are considered to contribute to the existing literature and to provide guidance for future research.
