In the search for novel therapeutic agents for Glioblastoma multiforme (GBM) treatment, anti-cancer peptides stand out due to their high selectivity potential. This study aims to investigate the in vitro efficacy and mechanisms of action of peptide fractions from Leiurus abdullahbayrami scorpion venom, which is a species with limited prior anti-tumoral research, using the C6 rat glioma cell line. Crude venom was separated via RP-HPLC using a gradient elution method. MTT cytotoxicity assays revealed that the major fraction collected at 30.2 minutes, designated as LabVT30.2, exhibited the highest potency. The IC50 value was calculated as 250 μg/mL (25 μg/100 μL) for a 48-hour treatment. At this concentration, cell migration, invasion, and tumorigenicity, which represent the aggressive hallmarks of GBM, were significantly suppressed at the 48th hour. AO/EB dual staining demonstrated that LabVT30.2 directed cells toward apoptosis, and flow cytometric analyses supported this process by showing a significant accumulation in the Sub-G1 population along with a decrease in the G1 phase. RT-qPCR analysis indicated that these effects were associated with the suppression of the WNT signaling pathway and the transcriptional activation of genes belonging to hem intrinsic and extrinsic apoptotic pathways. These findings were molecularly validated by a decrease in PCNA protein levels and an increase in total caspase-3 levels via in situ immunofluorescence staining. Furthermore, the elevation in active caspase-3 protein levels determined by flow cytometry provided additional molecular confirmation of this apoptotic induction. In conclusion, the LabVT30.2 fraction suppressed proliferation in C6 cells and induced cell cycle arrest through the down-regulation of Cyclin D1 expression via WNT pathway inhibition. By triggering apoptosis, it exhibited a potent anti-cancer profile. These data suggest that LabVT30.2 is a promising peptide-based candidate for GBM treatment and provides a foundation for further purification and structural analysis of the active peptide component.
