Özet
Toksoplazmoz, immün sistemi baskılanmış hastalarda ve AIDS?lilerde ciddi klinik tablolara neden olmaktadır. Atovakuon yan etkileri az olan bir antiprotozaoldır. Astragalus memebranaceus kök ekstresinin (AmE) immüniteyi stimüle etme etkisi ve fagositer hücrelerin aktivitesini arttırdığı, enfeksiyonlarda immünmodülatör etkiye sahip olduğu gösterilmiştir.Bu çalışmada; deneysel olarak Toxoplasma gondii ile enfekte edilen farelerde, atovakuon ve AmE tek tek ve kombinasyon olarak kullanılarak tedaviye ve IL-2, IL-12, INF gamma sitokin seviyelerine etkisinin olup olmadığının araştırılması amaçlanmıştır. Bu amaçla her biri sekiz BALB/c fareden oluşan dört deney grubu oluşturulmuş ve tüm fareler 2x10 üzeri 4/ml T.gondii RH suşu trofozoiti ile intraperitoneal yolla enfekte edilmiştir. 24 saat sonra, kontrol (K) grubu dışında, Atovakuon (Ato) grubuna atovakuon, Astragalus (Astra) grubuna AmE ve Ato+Astra tedavi grubuna ise atovakuon ve AmE birlikte oral gavaj yolu ile verilmiştir. Sekizinci günde tüm farelerden alınan periton sıvıları Thoma lamında sayılarak trofozoit sayıları belirlenmiş ve serum örneklerinin IL-2, IL-12, INF gamma düzeyleri ELISA ile araştırılmıştır. Toxoplasmosis is cause of serious clinical situations on immunspressed patients. Atovaquone is an antiprotozoal with a little side effect. Astragalus membranaceus root extract (AmE) has been showed as phagocytic activity enhancer, immunomodulator and immunstimulant agent. The aims of this study were to investigate the effectiveness of AmE alone and in combination with atovaquone and determine the levels of IL-2, IL-12 and INF gamma in experimentally infected mice with Toxoplasma gondii. Four experimental groups each consisting of eight BALB/c mice were infected intraperitoneally with 2x10 4/ml of T.gondii RH strain trophozoite. After 24 hours, atovaquone was given Atovaquon (Ato) group, AmE was given Astragalus (Astra) group, atovaquone plus AmE to Ato+Astra therapy group by oral gavage.On the 8th day, number of trophozoites in peritoneal fluid and the level of IL-2, IL-12, INF gamma in serum were determined by Thoma slide and ELISA, respectively.
