Özet
Estrogens, because of their antihormonal effects are extensively used in cancer therapy of male genital organs; having antiandrogenic effects, estrogens are used in the treatment of prostat cancer. However, this case mostly causes feminisation in the men. In our study, our aim is to examine the effects and reasons of ethynyl estradiol on testis germ cells, which is commonly used in this kind of treatment. For this study, we used 200-250gr, three and a half months old, male Wistar rats we kept under the same biological and physiological conditions.For two months we injected the first experiment group every two days and the second experiment group everyday 0.0005mg/kg ethynyl estradiol into intraperitoneal way. At the end of the second month we processed testis biopsy materials we got from every three groups for light and electron microscopic observations. Compared testis biopsy materials of the treated rats to the controls groups testis biopsy materials: a/In the testis of first group rats; a decrease in the number of germ cells layer, vacuoles in cytoplasm of Sertoli cells, an increase in the vesicules of smooth endoplasmic reticulum, hydratation in mitochondria were observed. There was a thickening of tubule basement membrane. As for germ cells, there was a decrease in the number of dark spermatogonia but there wasn't an important change in the number of light spermatogonia and primary spermatocyte. There was a decrease in the number of secondary spermatocyte, spermatid and spermium. We observed a degeneration in Leydig cells. 49b/ In the testis of the second group rats; we observed a shrink in diameter and irregularity in contour of tubules. We observed the absolute disappearance of dark spermatogonia, only the existence of light spermatogonia and primary spermatocyte and the disappearance of secondary spermatocyte, spermatid and spermium on the tubule epithelium. We observed an increasing degeneration of Sertoli cytoplasm, a ruin of Leydig cells. We also observed that Leydig cells replaced with connective tissue. Acccording to our findings, the dose of ethynyl estradiol caused ultrastructural defects, a degeneration in the Leydig cells, leading into the low of testosteron level in addition to this, it direct affected both Sertoli cells which supported germ cells and directly germ cells. For this reason, we couldnt see germ cells except light spermatogonium and primary spermatocyte on tubule epithelium. 50
