Özet

Amaç: Gaz sensörlerinde, pillerde, katalitik organik dönüşümlerde, elektro katalizde, fotokatalizde, güneş pillerinde, yakıtlarda, ısı transfer sıvılarında yaygın bir kullanıma sahip olan CuO nanopartikülleri oksidatif stres oluşturarak canlı hücrelerde toksik etkide bulunmaktadır. Resveratrol oksidatif stres ile mücadele eden bir antioksidandır. 2 boyutlu hücre kültürleri toksisite çalışmalarında önemli olmakla birlikte in vivo hücreleri taklitte yetersiz kalmakta ve sonuçlarının uygulanabilirliği sınırlanmaktadır. 3 boyutlu hücre kültür çalışmaları ise in vivo hücreleri taklit etmede oldukça başarılıdır. Bu çalışma CuO nanopartiküllerinin 2D RTG-2 hücrelerindeki ve 3D RTG-2 spheroidlerindeki toksik etkisine karşı resveratrolün koruyucu etkisi sitotoksisite, biyokimyasal ve genetoksisite açısından incelenmesini amaçlamıştır. Yöntem: 2D RTG-2 hücrelerinde ve 3D RTG-2 spheroidlerinde CuO nanopartiküllerinin LD50 dozları ve resveratrolün toksik olamayan dozları 24 ve 48 saatlik maruziyetleri için tespit edilmiştir. Daha sonra çift doz uygulamaları yapılarak resveratrolün en etkin koruyucu dozu belirlenmiştir. Burada sulforhodamin B (SRB) testi, Tripan Mavisi ile boyama yöntemleri kullanılmıştır. 2D ve 3D RTG-2 hücre kültürlerinde uygulamalar yapıldıktan sonra total RNA ekstraksiyonu, cDNA sentezi ve antioksidan enzim genlerinin (sod1, sod2, cat, gsr, gpx1a, and gpx4b), büyümeyle ilişkili genlerin (gh1, igf1, igf2), üreme geninin (bol) ve glikolitik enzim geninin (gapdh) ekspresyon seviyelerindeki değişimlerin tespiti için Real-Time PCR uygulamaları yapılmıştır. Kaspaz 3/7 aktivite tayini yapılmıştır. Genotoksik etkiler için comet testi yapılmıştır. Bulgular: CuO nanopartikülünün LD50 dozları 2D RTG-2 hücreleri için 24 ve 48 saatlik maruziyette 12.5 µg/mL olarak tespit edilirken 3D RTG-2 spheroidlerinde 24 saatlik maruziyette 75 µg/mL, 48 saatlik maruziyette 100 µg/mL olarak belirlenmiştir. Resveratrolün en etkin koruyucu dozu 2D RTG-2 hücrelerinde 24 ve 48 saatlik maruziyette sırasıyla 0.5 µM ve 0.75 µM olarak tespit edilmiştir. 3D RTG-2 spheroidlerinde ise bu değerler sırasıyla 1 µM ve 0.25 µM olmuştur. Real Time PCR çalışmalarına göre 2D RTG-2 hücrelerindeki ekspresyon verileri 3D RTG-2 spheroidlerindeki sonuçlarla tutarlı olmamıştır. Tüm uygulama gruplarındaki kaspaz 3/7 aktiviteleri, 48 saatlik maruziyetten sonra 2D RTG-2 hücrelerinde ve 3D RTG-2 sferoidlerinde tutarlılık göstermiştir. CuO NP uygulaması kontrol grubuna kıyasla genotoksik etkilere sahip olmuştur ve resveratrol uygulaması bu genotoksik etkileri hem 2D RTG-2 hücrelerinde hem de 3D RTG-2 sferoidlerinde 24 ve 48 saat maruziyette azaltmıştır. Sonuç: 2D RTG-2 hücrelerinde ve 3D RTG-spheroidlerinde CuO nanopartiküllerinin oksidatif stres aracılı toksisitesinde resveratrolün antioksidan etkinliği sitotoksisite, biyokimyasal ve genotoksik testler ile karşılaştırılmalı olarak gösterilmiştir. 2D RTG-2 hücrelerinden elde edilen verilerin 3D RTG-2 spheroidlerinden elde edilen sonuçlarla farklılığı ortaya konmuştur.

Purpose: CuO nanoparticles, which are widely used in gas sensors, batteries, catalytic organic conversions, electrocatalysis, photocatalysis, solar cells, fuels, and heat transfer fluids, have a toxic effect on living cells by creating oxidative stress. Resveratrol is an antioxidant that fights oxidative stress. Although 2-dimensional cell cultures are important in toxicity studies, they are insufficient to mimic in vivo cells and the applicability of their results is limited. 3D cell culture studies are very successful in mimicking the in vivo cells. This study aimed to investigate the protective effect of resveratrol against the toxic effect of CuO nanoparticles on the 2D RTG-2 cells and the 3D RTG-2 spheroids in terms of cytotoxicity, biochemical and genotoxicity. Method: LD50 doses of CuO nanoparticles and non-toxic doses of resveratrol were determined in the 2D RTG-2 cells and the 3D RTG-2 spheroids for 24 and 48 h of exposure. Then, double dose treatments were made and the most effective protective dose of resveratrol was determined. Here, Sulforhodamine B and Trypan Blue staining methods were used. After applications in the 2D and 3D RTG-2 cell cultures, total RNA extraction, cDNA synthesis, and Real-Time PCR studies were carried out for determining changes in the expression levels of antioxidant enzyme genes (sod1, sod2, cat, gsr, gpx1a, and gpx4b), growth-related genes (gh1, igf1, igf2), reproductive gene (bol) and glycolytic enzyme gene (gapdh). The Caspase 3/7 activity test was examined. The Comet assay was performed for genotoxic effects. Findings: The LD50 doses of CuO nanoparticles were determined as 12.5 µg/mL for the 2D RTG-2 cells at 24 h and 48 h of exposure, while 75 µg/mL at 24 h of exposure and 100 µg/mL at 48 h of exposure for the 3D RTG-2 spheroids. The most effective protective dose of resveratrol was found to be 0.5 µM and 0.75 µM in 2D RTG-2 cells at 24 and 48 hours of exposure, respectively. In the 3D RTG-2 spheroids, these values were 1 µM and 0.25 µM, respectively. According to Real-Time PCR studies, expression data in the 2D RTG-2 cells were not consistent with results in the 3D RTG-2 spheroids. The caspase 3/7 activities in all treatment groups were consistent in the 2D RTG-2 cells and the 3D RTG-2 spheroids after 48 h of exposure. CuO NPs treatment had genotoxic effects compared to the control group, and resveratrol treatment reduced these genotoxic effects in both the 2D RTG-2 cells and the 3D RTG-2 spheroids at 24 and 48 h of exposure. Results: The antioxidant activity of resveratrol in the oxidative stress-mediated toxicity of CuO nanoparticles in the 2D RTG-2 cells and the 3D RTG-2 spheroids was demonstrated in comparison with cytotoxicity, biochemical, and genotoxic tests. The data obtained from the 2D RTG-2 cells differed from the results obtained from the 3D RTG-2 spheroids.
