Özet
Endometriyum kanseri (EK), dünya çapında insidansı hızla artan bir kanser tipidir. Konvansiyonel kemoterapi tedavilerinin sınırlı başarısı sebebiyle kanser tedavisine yönelik çalışmalar hedef proteinlerin inhibisyonu üzerine yoğunlaşmıştır. PI3K/AKT/mTOR yolağı neredeyse tüm kanser tiplerinin ilerlemesinden sorumludur. Rapamisin, yolağa ait mTOR proteinini inhibe ettiği bilinen ve klinik olarak denenmiş bir bileşiktir. Bu tez çalışmasında MVB (1-8) olarak kodlanan sekiz farklı özgün bileşiğin PI3K/AKT/mTOR yolağı proteinleri üzerinde inhibe edici potansiyeli moleküler docking analizleri sonucunda sentezlendi. İn silico analizler sonrasında bileşiklerin, iki boyutlu ve sferoid olarak kültürlenmiş EK hücre hattı üzerindeki sitotoksik etkileri in vitro olarak analiz edildi. Rapamisin ile karşılaştırılan sonuçlarda etki potansiyelleri tespit edilen MVB-1, MVB-4 ve MVB-6 bileşiklerinin PI3K/AKT/mTOR yolağı proteinleri üzerindeki inhibice edici etkileri Western blot ve immünohistokimyasal analizlerle gösterildi. Bileşiklerin tahmini toksisite ve farmakokinetik potansiyelleri web tabanlı araçlar kullanılarak belirlendi. İki boyutlu kültürlerdeki analiz sonuçları, MVB-4 ve MVB-6 bileşiklerinin PI3K ve mTOR proteinlerini inhibe etmede başarılı olduğu ortaya konuldu. Sferoid kültürlerde MVB-4 bileşiğinin PI3K ve AKT1 proteinlerini inhibe edebildiği, mTOR proteini üzerinde ise anlamlı bir inhibisyon etkisi gösteremediği tespit edildi. MVB-6 bileşiği ise sadece mTOR proteinini inhibe edebildi. İn vitro ve in silico sonuçlar değerlendirildiğinde MVB-4 ve MVB-6 bileşiklerinin ilerleyen aşamalarda in vivo kanser modellerinde araştırılma potansiyellerinin olduğu öngörülmektedir.

Endometrial cancer (EC) is a type of cancer with a rapidly increasing incidence worldwide. Due to the limited success of conventional chemotherapy treatments, studies for cancer treatment have focused on the inhibition of target proteins. The PI3K/AKT/mTOR pathway is responsible for the progression of almost all cancer types. Rapamycin is a clinically tested compound known to inhibit the mTOR protein of the pathway. In this thesis study, the inhibitory potential of eight different unique compounds encoded as MVB (1-8) on PI3K/AKT/mTOR pathway proteins was synthesized as a result of molecular docking analysis. After in silico analysis, the cytotoxic effects of the compounds on the two-dimensional and spheroidally cultured EC cell line were analyzed in vitro. Inhibitory effects of MVB-1, MVB-4 and MVB-6 compounds, whose effect potentials were determined in the results compared with rapamycin, on PI3K/AKT/mTOR pathway proteins were demonstrated by Western blot and immunohistochemical analyzes. The estimated toxicity and pharmacokinetic potentials of the compounds were determined using web-based tools. Analysis results in two-dimensional cultures revealed that MVB-4 and MVB-6 compounds were successful in inhibiting PI3K and mTOR proteins. It was determined that MVB-4 compound was able to inhibit PI3K and AKT1 proteins in spheroid cultures, but did not show a significant inhibition effect on mTOR protein. MVB-6 compound was able to inhibit only mTOR protein. When in vitro and in silico results are evaluated, it is predicted that MVB-4 and MVB-6 compounds have the potential to be investigated in in vivo cancer models in the future.
