Gruplar Kontrol, pGlu-Val-Asp-Phe (5 mg/kg), MK-801 (0.15 mg/kg), pGlu-Val-Asp-Phe (5 mg/kg) + MK-801 (0.15 mg/kg) şeklinde oluşturulmuştur. Tetrapeptid açık alan testinde MK-801'in etkisini anlamlı şekilde tersine çevirdi (p<0.01). Sonuçlar: MK-801 farelere akut olarak uygulandığında antidepresan, anksiyolitik, hiperaljezik ve lokomosyon artırıcı etkiler göstermiştir. pGlu-Val-Asp-Phe farelerde 5 mg/kg dozunda anksiyolitik etki göstermiştir. pGlu-Val-Asp-Phe MK-801 ile kombine edildiğinde bazı testlerde MK-801'in etkisini kısmen artırırken, bazı testlerde değiştirmemiş, bazı testlerde ise anlamlı olarak tersine çevirmiştir. İlerleyen çalışmalarda daha spesifik in vitro testler kullanılarak pGlu-Val-Asp-Phe'in NMDA reseptörleri üzerindeki net etkisi aydınlatılmalıdır.
Özet (Çeviri)
Effect of Pyroglutamic acid-Valine-Aspartate-Phenylalanine (pGlu-Val-Asp-Phe) on depression, anxiety and analgesy in MK-801 administered mice Objective: Neurosecretory cells in the corpus cardiacum of insects synthesis some hormones called as adipokinetic, hypertrehaloasemic and hyperprolinaemic. In previous studies, AKH showed antidepressant effects in mice when administered chronically, increased neurotrophic factors and neurogenesis. In studies, it was shown that metabolites starting with pyroglutamic acid (pGlu) which were called as pyroglutamyl peptides were responsible from effects of AKH. Pyroglutamic acid-Valine-Aspartate-Phenylalanine (pGlu-Val-Asp-Phe) is a tetrapeptide structured pyroglutamyl peptide. In previous studies, pGlu-Val-Asp-Phe exerted antidepressant, partial anxiolytic and analgesic effects when administered acutely to mice. MK-801 is a NMDA receptor antagonist drug which is used to form experimental schizophrenia model in rat and mice. Effects of pyroglutamic acid and pyroglutamyl peptides on NMDA receptors were not showed in previous studies. Method: In this study, we investigated effects of pGlu-Val-Asp-Phe on depression, anxiety and analgesy in MK-801 administered mice using forced swimming, elevated plus maze and hot plate test. We used 9 weeks old male BALB/c mice weighted 25-35 g. Groups were control, pGlu-Val-Asp-Phe (5 mg/kg), MK-801 (0.15 mg/kg), pGlu-Val-Asp-Phe (5 mg/kg) + MK-801 (0.15 mg/kg). The number of the mice in each group was 8-10. Locomotor activity assessed by using open field test to avoid from false positive and negative results. Drugs were administered 30 minute before the behavioural tests. Results: MK-801 0.15 mg/kg significantly shortened immobility time in the forced swimming test when given alone or administered in combination with the tetrapeptide (p<0.001). MK-801 significantly increased % open arm time and % open arm entries in the elevated plus maze test (EPM) when given alone or administered in combination with the tetrapeptide (p<0.001). pGlu-Val-Asp-Phe significantly increased % open arm time (p<0.05) and % open arm entries (p<0.01) in the EPM test. MK-801 significantly decreased first licking time in the hot plate test when given alone or administered in combination with the tetrapeptide (p<0.001). MK-801 significantly increased total number of movements in the open field test when given alone (p<0.001) or administered in combination with the tetrapeptide (p<0.01). Tetrapeptide significantly reversed effect of MK-801 in the open field test (p<0.01). Conclusions: MK-801 exerted antidepressant, anxiolytic, hyperalgesic and locomotion enhancing effects when given acutely to mice. pGlu-Val-Asp-Phe showed anxiolytic effect at the dose of 5 mg/kg. pGlu-Val-Asp-Phe when combined with MK-801 partially increased effect of MK-801 in some tests while didn't alter the effect of MK-801 in some tests, and in some other tests significantly reversed the effect. Further studies using more spesific in vitro tests should be done to enlighten the clear effect of pGlu-Val-Asp-Phe on NMDA receptors.
