Özet

Bu tez çalışmasında, Samsun ilinde farklı satış noktalarından temin edilen açık ve kapalı formdaki bitki çayı örneklerinin mikrobiyolojik kalitesi değerlendirilmiştir. Çalışmada toplamda 15 farklı türde (adaçayı, ıhlamur, kekik, melisa, papatya, rezene, kuşburnu, lavanta, zencefil, ekinezya, anason, zencefil, defne, hatmi ve sinemaki) 150 bitki çayı örneği materyal olarak kullanılmıştır. Analiz kapsamında toplam mezofilik aerobik bakteri (TMAB), maya-küf, Bacillus cereus, Escherichia coli ve Salmonella spp. varlığı araştırılmıştır. Şüpheli bulunan izolatlar, tür düzeyinde tanımlama amacıyla MALDI-TOF MS ve PCR ile analiz edilmiştir. İdentifikasyonu yapılan B. cereus izolatlarında bazı toksin genleri (nheA, nheB, nhec, hblA, hblC, hblD, cytK1) ile toksin geni içeren suşlarda fenotipik antibiyotik direnç profili ortaya konmuştur. Elde edilen bulgulara göre, TMAB sayısı açıkta satılan çaylarda ortalama 3.97 log kob/g, kapalı çaylarda ise 4.41 log kob/g düzeyinde saptanmıştır. Maya ve küf sayısı açıkta satılan çaylarda sırasıyla ortalama 1.37 log kob/g ve 1.81 log kob/g, kapalı çaylarda ise 0.82 log kob/g ve 1.49 log kob/g olarak belirlenmiş olup istatistiksel olarak fark bulunmamıştır (p>0.05). Örneklerin tamamında E. coli saptama sınırının altında (2<log kob/g) değerlendirilmiş olup Salmonella spp.'ye ise rastlanılmamıştır. Toplam 150 bitki çayı örneğinin 20'sinde (20/150-%13,3) B. cereus varlığı ortaya konmuştur. Örnek dağılımında; açık bitki çayı örneklerinin 7'sinde (7/75-%9,3), kapalı çay örneklerinin ise 13'ünde (13/75-%17,3) etken tespit edilmiştir. İdentifikasyonu yapılan toplam 20 B. cereus izolatının 14'ünde (14/20-%70) en az bir toksin geni saptanmış olup, açık bitki çayı örneklerinde en fazla hblA geni (%42,9), kapalı örneklerde ise nheA (%61,5) geni pozitif bulunmuştur. nheA ve hblA gen varlığının örnek açısından istatistiksel farkı gözlenmiştir (p<0.05). Toksin gen pozitifliği gösteren 14 B. cereus izolatı üzerinde yapılan antibiyotik dirençlilik testleri sonucunda, Ampisilin direnci kapalı ürün orijinli izolatlarında daha yüksek (%87,5-%16,7; p<0.05). Klindamisin direnci ise açık ürün izolatlarında daha yüksek saptanmıştır (%50-%12,5; p<0.05). Bu çalışma, bitki çaylarının mikrobiyolojik açıdan risk taşıdığı örneklerdeki B. cereus varlığı ve etkendeki toksin genleri ile antibiyotik direnç özelliklerinin gıda güvenliği temelinde halk sağlığı açısından dikkate alınması gereken bir durum olduğu, ambalajlama ve hijyen koşullarının iyileştirilmesinin tüketici sağlığı açısından önem arz ettiğini sonucuna varılmıştır.

In this thesis study, the microbiological quality of herbal tea samples in packed and unpacked forms obtained from different sales points in Samsun province was evaluated. In the study, a total of 150 herbal tea samples of 15 different types (sage, linden, thyme, lemon balm, chamomile, fennel, rosehip, lavender, ginger, echinacea, anise, ginger, laurel, marshmallow and sennaki) were used as material. Within the scope of the analysis, the presence of total mesophilic aerobic bacteria (TMAB), yeast-mold, Bacillus cereus, Escherichia coli and Salmonella spp. was investigated. The isolates found to be suspicious were analyzed by MALDI-TOF MS and PCR to achieve species-level identification. In the identified B. cereus isolates, some toxin genes (nheA, nheB, nheC, hblA, hblC, hblD, cytK1) were detected, and phenotypic antibiotic resistance profiles were determined in the strains carrying these toxin genes. According to the obtained findings, TMAB count was determined as 3.97 log cfu/g on average in teas sold in the unpacked and 4.41 log cfu/g in packaged teas. Yeast and mold counts were determined as 1.37 log cfu/g and 1.81 log cfu/g on average in teas sold in the open, and 0.82 log cfu/g and 1.49 log cfu/g in packaged teas, respectively, and no statistically significant difference was found (p<0.05). E. coli was evaluated below the detection limit (2<log cfu/g) in all samples and Salmonella spp. was not detected. The presence of B. cereus was revealed in 20 of the 150 herbal tea samples in total (20/150-13.3%). In the sample distribution; The agent was detected in 7 (7/75-9.3%) of the unpacked herbal tea samples and in 13 (13/75-17.3%) of the packed tea samples. At least one toxin gene was detected in 14 (14/20-70%) of the 20 identified B. cereus isolates, and the most positive gene was hblA gene (42.9%) in unpacked herbal tea samples, while nheA (61.5%) gene was found positive in packed samples. A statistical difference was observed in the presence of nheA and hblA genes in terms of sample (p<0.05). As a result of antibiotic resistance tests performed on 14 B. cereus isolates showing toxin gene positivity, Ampicillin resistance was higher in isolates of packed product origin (87.5%-16.7%; p<0.05). Clindamycin resistance was found to be higher in unpacked product isolates (50%-12.5%; p<0.05). This study concluded that herbal teas carry a microbiological risk, the presence of B. cereus in the samples and the toxin genes in the agent and antibiotic resistance characteristics are a situation that should be taken into consideration in terms of public health on the basis of food safety, and that improving packaging and hygiene conditions is important for consumer health.
