Özet

Silene L. cinsi Caryophyllaceae familyasına ait olup Türkiye'de 148, Dünya'da ise 700'den fazla tür ile geniş bir yayılım göstermektedir. Etnobotanik kayıtlar farklı Silene türlerinin gıda veya tedavi amacıyla kullanıldığını göstermektedir. Silene L. cinsinin çok çeşitli sekonder metabolit içerdiği ve birçok farklı biyolojik aktiviteye sahip olduğu bildirilmiştir. Fenolik bileşikler ve flavonoitler gibi sekonder metabolitlerin antioksidan ve antienflamatuar etkileri, Silene L. cinsinde yaygın olarak bulunan fitoekdisteroitlerin ise adaptojenik etkileri sayesinde saç-kıl uzaması üzerinde destekleyici rol oynayabileceği bildirilmektedir. Bu doğrultuda tez çalışmamızda Silene dichotoma Ehrh. subsp. dichotoma türünün kimyasal içeriği ve biyolojik etkileri araştırılmıştır. Fitokimyasal çalışmalar, DPPH ve SO radikallerine karşı süpürücü etkinlikleri ile toplam fenol ve toplam flavonoit içerikleri sonucuna göre yönlendirilmiştir. Ekstre ve fraksiyonların sekonder metabolit profili LC/MS yöntemiyle belirlenmiştir. Fitokimyasal çalışmalar sonucunda 3 bileşiğin yapısı kersetin, izoramnetin ve izoramnetin 3-O-galaktozit olarak tanımlanmıştır. Ekstre ve fraksiyonların RAW 264.7 ve HaCaT hücrelerinde hücre canlılığı üzerindeki etkileri değerlendirilmiştir, HaCaT hücrelerinde H2O2 ile oluşturulan oksidatif hasara karşı koruyucu etkileri ve LPS ile uyarılmış RAW 264.7 hücrelerinde NO düzeyleri üzerinden antienflamatuar etkileri araştırılmıştır. Sonuçlar, ekstre ve fraksiyonların hücreler üzerinde belirgin bir sitotoksik etki göstermediğini ve antienflamatuar etkiye sahip olduğunu ortaya koymuştur ancak H2O2 kaynaklı hücresel hasarı n-hekzan alt fraksiyonu dışında önleyemediği saptanmıştır. Hücresel düzeydeki değişimlerin biyolojik mekanizmalarla ilişkisinin anlaşılması amacıyla HaCaT hücre hattında metabolomik çalışmalar da yürütülmüştür. Elde edilen bulgular doğrultusunda, S. dichotoma Ehrh. subsp. dichotoma türünün fitokimyasal içeriği, biyolojik potansiyeli ve hücresel düzeydeki etkileri belirlenmiştir.

The genus Silene L. belongs to the family Caryophyllaceae and shows a wide distribution with 148 species in Turkey and more than 700 species worldwide. Ethnobotanical records indicate that various Silene species have been used for food or medicinal purposes. The genus Silene L. has been reported to contain a wide variety of secondary metabolites and to exhibit numerous biological activities. It has been reported that the antioxidant and anti-inflammatory effects of secondary metabolites such as phenolic compounds and flavonoids, together with the adaptogenic effects of phytoecdysteroids commonly found in the genus Silene L., may play a supportive role in hair growth. Accordingly, in this thesis study, the chemical content and biological effects of Silene dichotoma Ehrh. subsp. dichotoma were investigated. Phytochemical studies were guided by the radical scavenging activities against DPPH and SO radicals, as well as total phenolic and total flavonoid contents. The secondary metabolite profile of the extracts and fractions was determined by LC/MS. As a result of the phytochemical studies, the structures of 3 compounds were identified as quercetin, isorhamnetin, and isorhamnetin 3-O-galactoside. The effects of the extracts and fractions on cell viability were evaluated in RAW 264.7 and HaCaT cells; their protective effects against H2O2-induced oxidative damage in HaCaT cells and their anti-inflammatory effects via NO levels in LPS-stimulated RAW 264.7 cells were investigated. The results showed that the extracts and fractions did not exhibit a significant cytotoxic effect on the cells and possessed anti-inflammatory activity; however, except for the n-hexane subfraction, they were unable to prevent H2O2-induced cellular damage. Metabolomic studies were also conducted in the HaCaT cell line to better understand the relationship between cellular changes and biological mechanisms. Based on the findings obtained, the phytochemical content, biological potential, and cellular-level effects of S. dichotoma Ehrh. subsp. dichotoma were determined.
