Introduction: Many surgical techniques and medical or herbal drugs have been experimented in order to increase regeneration capacity of the liver and experiments are still in course. Therefore, we have aimed to examine the effects of silymarin, which is administrated to the rats implemented with IPC before hepatectomy during the experimental liver resection, on the liver regeneration. Materials and Methods: The study has been started after receiving consent, from Local Ethics Committee for Animal Experiments of Gazi University, dated 26/06/2015 and coded G.Ü.ET-15.047. In the study, 30 male Wistar-Albino rats have been used. On the rats in the sham group, only hepatic manipulation and portal triad dissection have been implemented; and they have been sacrificed after 48 hours. On Group II (Partial hepatectomy), 70% liver resection has been implemented. Resected liver tissue has been weighed. Rats have been sacrificed after 48 hours. On Group III (Silymarin + Partial hepatectomy), 200 mg/kg of Silymarin dose have been implemented by adding it to their drinking water for six weeks. Six weeks later, 70% liver resection has been implemented on rats; and rats have been sacrificed after 48 hours. On Group IV (Ischemic preconditioning + Silymarin + Partial hepatectomy), Silymarin is administered for 6 weeks before the process, and then 70% liver resection has been implemented after ischemic preconditioning; and rats have been sacrificed after 48 hours. On Group V (Ischemic preconditioning + Partial Hepatectomy), after the ischemic preconditioning, 70% liver resection has been implemented; and rats have been sacrificed after 48 hours. After the hepatectomies implemented on rats, serum AST, LDH, Bilirubin, TNF-α and IL-6 values have been studied with ELISA method. Tissue samples have been histopathologically evaluated in terms of a number of mitosis, congestion, necrosis, cytoplasmic vacuolization and presence of neutrophils. PCNA (proliferating cell nuclear antigen) antibodies have been immunohistochemically studied. Results: In the group administered with Silymarin, it has been found out that regeneration capacity of remaining liver tissue had been higher, and liver functions had also been affected lesser compared to the group on which silymarin has not been administered. In addition to this, during the partial hepatectomy, it has been observed that IPC had increased regeneration rate of remaining liver tissue and liver functions had been affected lesser, and, at the same time, it has been found out that silymarin administration before hepatectomy had increased the regeneration rate much more and affected liver functions lesser. Conclusion: With these results, it can be said that silymarin administration and IPC implementation before hepatectomy would increase liver regeneration rate. However, in order for these results should be implemented in the clinic, similar clinical studies are required.
