Özet
HİPERTANSİYON TEDAVİSİNDE ORAL KULLANIM İÇİN NANO İLAÇ TAŞIYICI SİSTEMLERİN GELİŞTİRİLMESİ VE DEĞERLENDİRİLMESİ ÖZET Amaç: Bu çalışmanın amacı günümüzde aritmi, anjina pektoris ve hipertansiyon tedavisinde kullanılan nadolol (NDL) etkin maddesinin biyoyararlanım profilini iyileştirmek ve piyasada bulunan preparat formlarına alternatif oluşturmak amacıyla mikroemülsiyon, kendiliğinden emülsifiye ilaç taşıyıcı sistem (SEDDS) ve lipozom ilaç taşıyıcı sistemlerine yüklemek önformülasyon çalışmalarını gerçekleştirmektir. Gereç ve Yöntem: Lipozomlar etanol enjeksiyon yöntemi, mikroemülsiyon ve SEDDS'ler distile su ile titre edilerek psödo üçgen faz yöntemiyle hazırlanmıştır. Formülasyonların damlacık/partikül boyutu, polidispersite indeksi (PDI), zeta potansiyeli, iletkenlik, refraktif indeks, pH, viskozite, termodinamik stabilite, seyreltmeye dayanıklılığı, enkapsülasyon verimliliği, ilaç yükleme etkinliği, enzimatik degredasyona dayanıklılık, in vitro salım, hücre kültürü çalışmaları, uzun dönem stabilite ve in situ tek geçişli intestinal perfüzyon (SPIP) çalışmaları yapılmıştır. Sitotoksisite çalışmasında formülasyonlar %95'in üzerinde hücre canlılığı göstermiştir. SPIP çalışmasında geliştirilen mikroemülsiyon ve SEDDS'lerin NDL permeabilitesini artırdığı tespit edilmiştir. Sonuç: Geliştirilen nanotaşıyıcı sistemlerin NDL'nin biyoyararlanımını iyileştirdiği ve piyasadaki preparatlarına alternatif oluşturabileceği düşünülmüştür.
Özet (Çeviri)
DEVELOPMENT AND EVALUATION OF NANO DRUG DELIVERY SYSTEMS FOR ORAL USE IN THE TREATMENT OF HYPERTENSION ABSTRACT Aim: The aim of this study was to improve the bioavailability profile of the active ingredient nadolol (NDL), which is currently used in the treatment of arrhythmia, angina pectoris and hypertension, and to perform pre-formulation studies to load it into microemulsion, self-emulsifying system (SEDDS) and liposome drug carrier systems in order to create an alternative to commercially available preparation forms. Materials and Methods: Liposomes were prepared via ethanol injection method, microemulsion and SEDDS were prepared via pseudo-triangular phase method by titrating with distilled water. Droplet/particle size, polydispersity index (PDI), zeta potential, conductivity, refractive index, pH, viscosity, thermodynamic stability, resistance to dilution, encapsulation efficiency, drug loading efficiency, resistance to enzymatic degradation, in vitro release, cell culture studies, long-term stability and in situ single pass intestinal perfusion (SPIP) studies were performed. Results: The droplet/particle size, PDI value and zeta potential of the developed microemulsions, SEDDS and liposomes were in agreement with the literature. Ideally selected microemulsions and SEDDSs were found to be thermodynamically stable. The viscosity and pH values of the microemulsions and SEDDSs were suitable for oral use. No precipitation or phase separation was observed in SEDDSs in the dispersion stability study. Microemulsions and SEDDSs were investigated under different ambient conditions during 12 months for long-term stability determination and no physical instability problems were encountered. In the study of enzymatic degradation, it was found that chitosan was able to protect liposomes in gastric environment. In the in vitro release study, microemulsion and SEDDS formulations showed 100% release in both media. Liposome formulations showed more controlled release. In cell studies, formulations showed a higher transition from apical to basolateral direction. In cytotoxicity study, formulations showed cell viability above 95%. In the SPIP study, it was found that microemulsions and SEDDSs developed increased NDL permeability. Conclusion: It is believed that the nanocarrier systems developed will improve the bioavailability of NDL and provide an alternative to marketed products.
