Objective: The aim of this study was to evaluate the effects of different manufacturing methods and thermal cycling on the mechanical properties of denture base materials. Materials and Methods: In our study, 4 groups (Vita Vionic (VV), Ivoclar Ivotion (IV), Yamahachi PMMA Disk (YA), Tempo CAD On Dent (OD)) produced by milling method from CAD/CAM (computer-aided design/computer-aided manufacturing) pre-polymerized disks, A total of 120 specimens were produced in 6 groups (n=20), including 1 group produced by printing on a three-dimensional (3D) printer (Alias 3D Printing Resin (3D)) and 1 group produced by the traditional heat polymerization method (Schütz Dental Futura Basic Heat (KN)) as a control group, and 12 subgroups (n=10) by applying thermal cycling to half of the specimens in all groups. The samples were subjected to 10,000 thermal cycles in a thermal cycling device (5 - 55°C). Flexural strength values were measured by three-point bending test on a Universal testing machine (Instron) and hardness values were measured by Vickers hardness tester. Fracture surfaces of the samples were evaluated by scanning electron microscopy (SEM). Data were analyzed using one-way analysis of variance (ANOVA) and Tukey HSD test (α = 0.05). Results: While 4 CAD/CAM milling groups (VV, IV, YA, OD) exhibited the highest flexural strength values in the group without thermal cycling (p<0.05), CAD/CAM milling groups except YA group also exhibited the highest flexural strength values in the groups with thermal cycling (p<0.05). The lowest flexural strength values in the groups without thermal cycling were seen in the KN and 3D group and the results were similar (p>0.05), while the lowest flexural strength values in the groups with thermal cycling were seen in the 3D group (p<0.05). In the groups without thermal cycling, the hardness values in the VV and 3B groups were similar among themselves (p>0.05), but significantly lower than all other groups (p<0.05) and the highest hardness values were seen in the OD group. In the groups where thermal cycling was applied, the hardness value of the 3B group was higher than the VV group and lower than all other groups (p<0.05). Conclusion: Denture base materials produced by CAD/CAM milling method exhibited high flexural strength values. The resin produced by 3D printing method showed significant differences in terms of mechanical properties with the samples in the CAD/CAM milling group except for the VV group. The VV group showed the lowest hardness value in all groups with and without thermal cycling, while the OD group showed the highest hardness value in the groups without thermal cycling. Thermal cycling only increased the flexural strength of the traditional denture base material group (KN).
