Özet

Bu çalışmanın amacı, pirinç, nohut ve kabak çekirdeği proteinlerinin hazır domates ve mercimek çorbalarının duyusal, fonksiyonel ve biyoaktif özelliklerini iyileştirme potansiyelini kapsamlı bir şekilde araştırmaktır. Proteinler, insan beslenmesinin temel yapı taşlarından biri olup, yalnızca besleyici değerleriyle değil, aynı zamanda emülsifikasyon, jelleşme, su ve yağ tutma gibi fonksiyonel özellikleriyle de gıda endüstrisinde yaygın olarak kullanılmaktadır. Günümüzde hayvansal proteinler, yüksek biyoyararlanım ve tam amino asit profili nedeniyle sıklıkla tercih edilse de, üretim süreçlerinin çevresel etkileri, sürdürülebilirlik kaygıları ve bazı sağlık riskleri, bitkilerden elde edilen alternatif protein kaynaklarına olan ilgiyi artırmıştır. Bitkisel proteinler; düşük üretim maliyetleri, çevre dostu üretim süreçleri ve sağlık açısından sundukları potansiyel faydalarla dikkat çekmektedir. Ancak bu kaynaklar, düşük çözünürlük, esansiyel amino asit profilleri ve bazı durumlarda istenmeyen tat özellikleri gibi dezavantajlara da sahiptir. Bu bağlamda, çalışmanın temel amacı; pirinç, nohut, bakla, bezelye, kabak çekirdeği proteinleri ile kurutulmuş yulaf şurubu olmak üzere altı farklı bitkisel kaynağın hazır domates ve mercimek çorbalarına eklenerek, bu proteinlerin, ürünlerin duyusal özellikleri, tüketici kabulü, biyoaktif ve fonksiyonel özellikleri üzerindeki etkilerini karşılaştırmalı olarak incelemektir. Çalışmada, çorba formülasyonlarına kuru ağırlıkça %10 oranında bitkisel protein kaynağı eklenmiş ve ardından gastrointestinal sindirimi simüle eden in vitro analizler gerçekleştirilmiştir. Bu analizlerde toplam fenolik içerik, antioksidan kapasite ve çözünürlük gibi biyoaktif parametrelerin yanı sıra, viskozite, akışkanlık ve köpük oluşturma gibi fonksiyonel özellikler de değerlendirilmiştir. Duyusal analizler sonucunda, altı bitkisel kaynak arasından pirinç, nohut ve kabak çekirdeği proteinleri en yüksek puanları alarak çalışmanın ileri aşamalarında detaylı incelemeye alınmıştır. Tat, aroma ve kıvam gibi duyusal parametreler üzerinden yapılan değerlendirmelerde, kabak çekirdeği proteini domates çorbası ile, nohut proteini ise mercimek çorbası ile en yüksek uyumu göstermiştir. Pirinç proteini ise nötr tadı ve pürüzsüz dokusuyla olumlu bir duyusal profil sunmuş, ancak düşük çözünürlük nedeniyle bazı matrislerde çökme eğilimi göstermiştir. Protein kalitesinin değerlendirilmesinde kullanılan PDCAAS (Protein Sindirilebilirliği Düzeltmeli Amino Asit Skoru) değerleri, bitkisel proteinlerin sınırlayıcı amino asitler nedeniyle hayvansal proteinlere kıyasla daha düşük olduğunu göstermektedir. Bu çalışmada, nohut proteini 84, kabak çekirdeği proteini 80 ve pirinç proteini 58 PDCAAS değeri ile değerlendirilmiştir. Mercimek çorbası matrisinde metiyonin, domates çorbasında ise valin ve lösin sınırlayıcı amino asitler olarak öne çıkmıştır. Bu durum, farklı bitkisel proteinlerin kombinasyonlarının amino asit tamamlayıcılığı açısından önemli bir potansiyel taşıdığını göstermektedir. Protein çözünürlüğü analizlerinde, nohut proteini en yüksek çözünürlüğü göstermiştir. Özellikle mercimek çorbası matrisinde kabak çekirdeği proteini %56,98 çözünürlük ile öne çıkarken, pirinç proteini domates çorbasında düşük çözünürlük sergilemiş ancak mercimek çorbasında daha iyi performans göstermiştir. Bu bulgular, ürün matrisinin pH'sı, bileşen etkileşimleri ve protein yapısının çözünürlük üzerinde belirleyici olduğunu ortaya koymaktadır. Antioksidan kapasite analizlerinde, kabak çekirdeği proteini eklenen örnekler hem domates hem de mercimek çorbasında en yüksek değerleri vermiştir. Bu sonuç, kabak çekirdeğinin doğal fenolik bileşenleriyle açıklanmakta ve bu proteinin fonksiyonel gıda formülasyonlarında antioksidan kapasiteyi artırmak için ideal bir bileşen olduğunu göstermektedir. Nohut ve pirinç proteinleri ise daha düşük antioksidan değerler sunmuştur. Viskozite ve akışkanlık ölçümleri, protein ilavesinin çorba matrislerinin fiziksel özelliklerini önemli ölçüde etkilediğini göstermiştir. Domates çorbası örnekleri, mercimek çorbasına kıyasla daha iyi akışkanlık göstermiştir. Ffc katsayısı değerlendirildiğinde, domates çorbası örnekleri“az yapışkan”sınıfında yer alırken, mercimek çorbası örnekleri“çok yapışkan”sınıfına yaklaşmıştır. Nohut proteini, yüksek lif ve nişasta içeriği nedeniyle zayıf akışkanlık göstermiştir. Fiziksel analizlerde, tüm örneklerin pH, nem ve su aktivitesi değerleri mikrobiyal stabilite açısından güvenli sınırlar içinde kalmıştır. Domates çorbası örnekleri asidik karakterini korurken, mercimek çorbası nötre yakın pH değerleri sergilemiştir. Düşük su aktivitesi değerleri, ürünlerin raf ömrünü uzatmakta ve mikrobiyal bozulmayı önlemektedir. Fonksiyonel analizlerde, nohut proteini yüksek yağ tutma kapasitesi ve orta düzey köpük stabilitesi ile öne çıkarken, kabak çekirdeği proteini düşük yağ tutma kapasitesi göstermiş ancak yüksek köpük stabilitesi ile dikkat çekmiştir. Bu durum, kabak çekirdeği proteininin daha hidrofobik yapısı ve düşük yüzey aktivitesi ile ilişkilidir. Sonuç olarak, bu tez çalışmasında elde edilen bulgular, bitkisel proteinlerin hazır çorba formülasyonlarında başarılı bir şekilde kullanılabileceğini göstermektedir. Nohut proteini, yüksek çözünürlük ve analiz verimi ile öne çıkarken; kabak çekirdeği proteini, antioksidan kapasitesi ve duyusal uyumu ile dikkat çekmiştir. Pirinç proteini ise bazı matrislerde düşük çözünürlük nedeniyle sınırlı performans göstermiştir. Ancak uygun ürün matrislerinde bu dezavantajlar minimize edilebilir. Protein kombinasyonları ile PDCAAS değerlerinin optimize edilebileceği ve fonksiyonel, besleyici, çevresel açıdan sürdürülebilir ürünlerin geliştirilebileceği sonucuna varılmıştır. Bu çalışma ile elde edilen bugular, bitkisel proteinler ile tüketici ihtiyaçlarını karşılayan, besleyici ve çevresel açıdan sürdürülebilir gıda ürünleri geliştirme konusundaki bilimsel ve endüstriyel yaklaşımlara önemli bir katkı sunmaktadır.

In response to the global shift toward sustainable, plant-based nutrition, this study investigates the integration of selected plant-based protein sources namely rice, chickpea, and pumpkin seed proteins into instant tomato and lentil soup formulations. The work presents a comparative evaluation of their impacts on the sensory attributes, functional characteristics, and bioactive potentials of these widely consumed convenience foods. The overarching aim is to determine the suitability of these plant proteins as viable alternatives to animal proteins in ready-to-eat products, considering both nutritional quality and consumer acceptability. Proteins of plant origin are increasingly favored for their economic feasibility, environmental friendliness, and health-promoting properties. However, compared to their animal-derived counterparts, they often exhibit structural and functional limitations such as low solubility, incomplete amino acid profiles, and potential sensory drawbacks. Addressing these challenges requires a detailed understanding of each source's compositional characteristics and performance within complex food systems. In the present study, protein isolates from six plant-based sources, rice (Oryza sativa), chickpea (Cicer arietinum), faba bean (Vicia faba), pea (Pisum sativum), pumpkin seed (Cucurbita pepo), and dried oat syrup, were initially screened by incorporating 10% (dry weight) into instant tomato and lentil soup matrices. Sensory evaluations were conducted to assess attributes such as taste, aroma, mouthfeel, color, and overall acceptability. Based on panelist feedback, rice, chickpea, and pumpkin seed proteins were selected for further analysis. Each of the selected protein sources exhibits a unique composition and functional profile. Chickpea protein contains approximately 18–24% protein with a well-balanced amino acid profile rich in lysine and tryptophan, but relatively deficient in methionine and cysteine. It is also known for its high dietary fiber and moderate starch content, which contribute to texture and satiety. Pumpkin seed protein, with a protein content of 30–40% depending on genotype and processing, is notable for its richness in leucine, arginine, and glutamic acid, along with a naturally high level of phenolic compounds such as gallic and caffeic acids. This composition grants pumpkin seed protein a strong antioxidant profile. Rice protein, derived primarily from the endosperm, offers up to 78% protein in isolate form and is rich in methionine and valine, though it contains lower levels of lysine, limiting its standalone nutritional completeness. Protein quality was evaluated using the PDCAAS (Protein Digestibility-Corrected Amino Acid Score), a metric that accounts for both essential amino acid content and digestibility. Chickpea protein scored 84, pumpkin seed protein 80, and rice protein 58. While these scores are lower than those of animal proteins, they demonstrate the potential for nutritional optimization through strategic combinations of complementary plant proteins. In terms of sensory compatibility, pumpkin seed protein proved most suitable for tomato soup, providing a balanced flavor and smooth mouthfeel that enhanced the natural acidity of the base. Chickpea protein was better suited to lentil soup, where its creamy texture and nutty flavor complemented the matrix. Rice protein offered a mild taste and soft texture across both matrices, though its low solubility, particularly in acidic environments like tomato soup, resulted in some sedimentation. Solubility testing showed chickpea protein to be the most soluble overall, followed by pumpkin seed protein (notably achieving 56.98% solubility in lentil soup). Rice protein had lower solubility, especially in tomato soup, due to matrix-pH interactions and inherent structural properties. Functional measurements such as viscosity and flow behavior further illustrated matrix-specific effects: lentil soups were significantly more viscous and adhesive than tomato soups, and chickpea protein contributed to reduced flowability due to its fiber and starch content. Antioxidant assays (including total phenolic content and CUPRAC/Trolox equivalent antioxidant capacity) confirmed the superior bioactivity of pumpkin seed protein, attributed to its natural polyphenol composition. This enhancement was maintained even after simulated gastrointestinal digestion. Chickpea protein yielded moderate antioxidant effects, while rice protein showed minimal activity in this respect. Physicochemical analyses demonstrated that all soup formulations maintained pH, moisture, and water activity values within microbiologically safe and shelf-stable ranges. Tomato soups remained acidic (pH < 4.5), while lentil soups had near-neutral pH, supporting microbial safety and flavor stability. Water activity values (<0.6) indicated reduced spoilage risk and extended product shelf life. Further functional assessments revealed that chickpea protein exhibited the highest oil-holding capacity, beneficial for mouthfeel and flavor retention, whereas pumpkin seed protein demonstrated superior foam stability, likely due to its hydrophobic amino acid composition and moderate surface activity. These properties contribute to the structural integrity and sensory appeal of the final product. In conclusion, this research validates the use of rice, chickpea, and pumpkin seed proteins as viable plant-based alternatives for enriching instant soup formulations. Chickpea protein offers favorable solubility, nutritional quality, and sensory integration in legume-based matrices such as lentil soup. Pumpkin seed protein contributes strong antioxidant potential and functional stability in acidic systems like tomato soup. Rice protein, despite its sensory neutrality and textural contribution, requires formulation adjustments to mitigate its solubility limitations. The study highlights the importance of selecting the right plant protein for each matrix and supports the strategic blending of protein sources to enhance amino acid completeness (PDCAAS), functionality, and consumer appeal. Overall, the findings underscore the potential of plant proteins in developing nutritious, shelf-stable, and sustainable ready-to-eat products. The incorporation of these proteins not only addresses modern dietary needs but also aligns with broader goals of environmental stewardship and food innovation. These insights not only expand the scientific understanding of plant protein integration in convenience foods but also open new avenues for product development in both functional and clean-label categories. As consumer awareness around health, sustainability, and dietary quality continues to rise, the development of nutritionally enriched, plant-protein-fortified instant soups could offer an accessible and scalable solution. Future research may explore the use of protein hydrolysates or fermentation-based modifications to further improve solubility and sensory performance, while also investigating protein interactions with other food components in multi-ingredient matrices. In this regard, the findings of this thesis provide a valuable foundation for advancing both academic inquiry and industrial innovation in the plant-based food sector.
