Özet
Endokannabinoid sistem, enerji dengesi, metabolizma, gıda alımı ve vücut homeostazının düzenlenmesinde rol oynayan endojen bir sinyal sistemidir. Bu sistem, spesifik reseptörler, bunların ligandları olan endokannabinoidler (EC) ve ilgili enzimlerden oluşmaktadır. Ayrıca kanser, kardiyovasküler hastalıklar, ruh hali bozuklukları, bellek kaybı ve şizofreni gibi çeşitli hastalıklarla ilişkilendirilmiştir. EC'ler, başlıca yağ asidi amidleri ve monoaçilgliseroller (MAG) sınıfına giren lipit bazlı moleküllerdir. En iyi bilinen örnekleri, N-araşidonoiletanolamid (anandamid, N-AEA) ve 2-araşidonoilgliserol (2-AG) olup, CB1 ve CB2 reseptörlerine bağlanarak aktive olurlar. Bunun yanı sıra, doymuş ve doymamış yağ asitlerinden türeyen N-açiletanolamidler (NAE) gibi endokannabinoid benzeri (ECL) bileşikler de tanımlanmıştır ve bunlar EC'lerin metabolik yıkımını engelleyerek benzer biyolojik etkiler gösterebilir. ECL grubuna, N-açilaminoasitler, N-açilnörotransmitterler ve birincil yağ asidi amidleri de dahildir. ECL bileşikler kannabinoid reseptörleriyle etkileşime girmez; GPR119 (G proteinine bağlı reseptör 119), TRPV1 (Transient reseptör potansiyel vanilloid 1) ve PPARα (Peroksizom proliferatör aktive edici reseptör α) gibi reseptörlerle etkileşerek kannabinoid benzeri etkiler gösterebilirler. İnsanlarda bulunan EC ve ECL bileşikler, gıdalarla alınan yağ asitlerinden sentezlenir; bu bağlamda çoklu doymamış n-6 ve n-3 yağ asitleri en önemli kaynakları oluşturur. Yapılan çalışmalar, diyet yağının türü veya miktarındaki değişimlerin EC ve NAE düzeylerini farklı dokularda modüle edebildiğini ve bunun sonucunda obezite ve kilo artışı gibi sağlık çıktıları üzerinde etkili olabileceğini göstermiştir. EC ve ECL bileşikler yalnızca insanlarda değil, bazı bitki ve mikroorganizma sistemlerinde de bulunur. Ancak farklı gıda matrislerindeki profilleri ve konsantrasyonları halen yeterince bilinmemektedir ve bu bileşiklerin gıdalardaki varlığı ve oluşumları hakkında literatürde sınırlı bilgi bulunmaktadır. Buradan yola çıkarak, bu tez kapsamında, farklı fermente gıdalarda (sucuk ve peynir) EC ve ECL bileşiklerinin tespit edilmesi, bu bileşiklerin oluşum ve dönüşüm mekanizmalarının aydınlatılması ve sindirim süreci boyunca uğradıkları biyokimyasal değişimlerin belirlenmesi amaçlanmaktadır. Bununla birlikte, bağışıklık sistemi, iştah regülasyonu, ağrı iletimi ve merkezi sinir sistemi işlevlerinde kritik roller üstlenen bu lipit türevi moleküllerin, farklı EC/ECL profiline sahip fermente gıda tüketen deney hayvanlarının davranışları üzerindeki etkilerinin değerlendirilmesi yoluyla, fermente gıdaların gıda alımı ve diyetle ilişkili duygudurum üzerindeki potansiyel etkilerinin aydınlatılması hedeflenmektedir. Tezin ilk kısmında, farklı sucuk örneklerinin EC/ECL bileşik profili analiz edilmiş ve bu örneklerin NAE, N-açilaminoasit ve 2-MAG grubu bileşikler bakımından zengin olduğu ortaya konmuştur. Ancak çoklu faktör analizi (MFA) sonucunda, fermente ve ısıl işlem uygulanmış sucuklar arasında işleme koşulları açısından anlamlı bir farklılık gözlenmemiştir. Elde edilen bulgulara göre, laboratuvar ölçeğinde üretilen sucuklarda fermantasyon işlemi N-açilaminoasit ve 2-MAG bileşikleri oluşumunu artırmıştır. Buna karşın, ısıl işlem NAE düzeylerinde artışa yol açarken, 2-MAG miktarlarını azaltmıştır. Laboratuvar ölçekli sucuk üretiminde uygulanan olgunlaştırma/kurutma işlemlerinin ise EC ve ECL bileşiklerinde genel olarak anlamlı bir değişime neden olmadığı görülmüştür. Fermente sucuklarda depolama süreci boyunca, N-açilaminoasitlerin konsantrasyonlarında belirgin bir artış gözlemlenirken, 2-MAG düzeyleri olumsuz yönde etkilenmiştir. 2-MAG grubu bileşiklerde azalma, bu bileşiklerin yapısal olarak kararsız olmaları nedeniyle 1-MAG izomerlerine dönüşmesi ile ilişkili olabilir. Bu bölüm, gıda işleme yöntemlerinin sucuklardaki EC ve ECL bileşiklerinin profili ve miktarları üzerindeki etkilerine ilişkin önemli bir bakış açısı sunmaktadır. Tezin ikinci kısmında, taze ve olgunlaştırılmış peynirlerin EC/ECL bileşik profilleri karşılaştırılmış ve olgunlaşma sürecinin lipit sinyal iletiminde rol alan bileşiklerin oluşumunda belirleyici olduğu belirlenmiştir. Özellikle Danish Blue gibi olgunlaştırılmış peynirlerin, olgunlaşma süresince gerçekleşen yoğun lipoliz ve proteoliz nedeniyle yüksek düzeyde N-açilaminoasit türevi bileşik içerdiği belirlenmiştir. Cheddar peynirinde olgunlaşma aşamasında N-oleoilfenilalanin, N-linoleoilfenilalanin, N-oleoillösin ve N-palmitoilglisin düzeylerinde artış saptanmıştır. Buna karşılık, laboratuvar koşullarında üretilen taze kaşar peynirinde depolama sırasında gerçekleşen parçalanma nedeniyle 2-oleoilgliserol miktarı önemli ölçüde azalmıştır. Ayrıca, yüksek yağ içeriği ve lipoliz etkisiyle 2-MAG grubu bileşiklerin sucuklarda peynir örneklerine kıyasla daha yüksek konsantrasyonlarda bulunduğu sonucuna ulaşılmıştır. Tezin bu kısmında yapılan çalışmalar fermantasyon, olgunlaşma ve depolamanın gıdalardaki EC/ECL bileşik profili üzerindeki etkilerine ve bu bileşiklerin oluşumuna ilişkin olası mekanizmalara dair bilgiler sunmaktadır. Tezin üçüncü kısmında, N-açilaminoasit grubu bileşiklerin gıdalardaki biyosentez yolları ve metabolik oluşum mekanizmalarını anlamak için model sistemler tasarlanmıştır. Bu bağlamda sucuk benzeri model sistemden elde edilen bulgular, mikroorganizmaların N-açilaminoasit grubu bileşiklerin oluşumunda etkili olabileceğini göstermiştir. Geliştirilmiş model besiyeri sisteminde de bu durum doğrulanmış; aminoasit (glisin), yağ ve starter kültür varlığında N-açilaminoasit grubu bileşiklerin (oleoilglisin ve palmitoilglisin) oluştuğu, kültür bulunmadığında ise oluşmadığı belirlenmiştir. Elde edilen sonuçlar, kültür kaynaklı olası bir N-açiltransferaz benzeri enzimin rolüne işaret etmektedir. Ayrıca 2-MAG bileşiklerinin eklenen yağda doğal olarak bulunduğu, sucuk fermantasyonu ile uyumlu şekilde model sistemde de miktarının arttığı; ancak besiyeri ortamında kültür varlığında 2-oleoilgliserol ve 2-linoleoilgliserol'ün azaldığı saptanmıştır. Bu farklılığın, besiyeri modelinde yalnızca kültür kaynaklı enzimlerin aktif olmasından; buna karşılık sucuk benzeri model sistemde hem mikrobiyal hem de etten gelen endojen lipolitik enzimlerin yanı sıra yüksek yağ içeriğinin de sürece katkı sağlamasından kaynaklandığı düşünülmektedir. Bu kısımdan elde edilen bulgular, N-açilaminoasit grubu bileşiklerin gıdalardaki varlığına ve oluşumuna etki eden endojen ya da mikrobiyal enzimatik aktivitelerin olası mekanizmalarına ışık tutmaktadır. Tezin dördüncü kısmında, fermente sucuk ve eski kaşar peyniri üzerinde gerçekleştirilen in vitro sindirim deneyleri, bu ürünlerin sindirim sürecinde EC ve ECL bileşikleri açısından önemli potansiyel kaynaklar olduğunu göstermiştir. Özellikle intestinal fazda N-açilaminoasit ve 2-MAG bileşiklerinde artış gözlenmiştir. Sindirim sırasında bu bileşiklerin düzeyindeki artışın, hem bu bileşiklerin yapıdan ayrılarak serbest hale gelmesi hem de yeni sentez süreçlerinden kaynaklanabileceği belirlenmiştir. Model sistemde yapılan in vitro sindirim deneyinin bulguları ise N-açilaminoasit grubu bileşiklerin oluşumunda öncül maddelerin (yağ asidi ve amino asit) belirleyici olduğunu ortaya koymuştur. Özellikle intestinal fazda yağ asidi ve amino asit varlığında bazı biyolojik açıdan anlamlı lipit türevi bileşiklerin sentezlenebileceği; bu süreçte sindirim enzimlerinin potansiyel katkısıyla yeni oluşum mekanizmalarının devreye girebileceği anlaşılmaktadır. Sonuç olarak, gıda bileşimi ve sindirim enzimlerinin bu biyoaktif lipitlerin profilini yönlendirerek potansiyel sağlık etkilerini şekillendirebileceği söylenebilir. Endokannabinoid sistem, iştah düzenlenmesi ve enerji dengesinin kontrolünde kritik bir rol oynamaktadır. Tezin son kısmında yapılan çalışmalar, peynir türlerinin farelerde gıda alımı ve davranışları etkileyebildiğini göstermiştir. Taze kaşar verilen farelerde daha yüksek gıda tüketimi gözlenmiş; bu durum, taze kaşarın EC/ECL profili ile özellikle 2-OG ile N-açilaminoasit bileşiklerini, eski kaşar peynirine kıyasla düşük düzeyde içermesiyle ilişkilendirilmiştir. Buna karşılık, eski kaşar peynirinin daha zengin EC/ECL bileşimi, eski kaşar verilen grupta iştah azaltıcı etkiler göstererek farelerde artan tokluk hissi ve daha düşük anksiyete benzeri davranışlarla ilişkilendirilmiştir. Bu bulgular, peynirlerin EC/ECL profillerinin enerji dengesi ve ruh hali üzerinde belirleyici rol oynayabileceğini ortaya koymaktadır. Sonuç olarak, bu tez çalışması, farklı işleme yöntemlerinin fermente gıdalardaki EC/ECL bileşik profili üzerindeki etkilerini ve oluşum mekanizmalarına dair olası süreçleri ortaya koymaktadır. N-açilaminoasit grubundaki bileşiklerin oluşumunda mikroorganizmaların kritik rol oynadığı ve ayrıca sindirim sırasında bu bileşiklerin ortaya çıkabileceği gösterilmiştir. Olgunlaşmış peynir tüketiminin ruh hali ve iştah üzerinde etkiler yaratabileceği de bu tez kapsamında belirlenmiştir. Bu bulgular, fermente gıdaların bileşimi ve işleme koşullarının sağlık üzerinde potansiyel etkileri olabileceğini vurgulamaktadır, ancak bu lipit türevi bileşiklerin biyoyararlanımı ve in vivo fizyolojik etkilerini net bir şekilde ortaya koymak için daha fazla araştırmaya ihtiyaç vardır.

The endocannabinoid system is an endogenous signaling system involved in the regulation of energy balance, metabolism, food intake, and body homeostasis. This system consists of specific receptors, their ligands, endocannabinoids (ECs), and related enzymes. It has also been linked to various diseases, including cancer, cardiovascular disease, mood disorders, memory loss, and schizophrenia. ECs are lipid-based molecules primarily belonging to the class of fatty acid amides and monoacylglycerols (MAGs). The most well-known examples are N-arachidonoylethanolamide and 2-arachidonoylglycerol, which activate CB1 and CB2 receptors by binding to them. Additionally, endocannabinoid-like (ECL) compounds, such as N-acylethanolamides (NAEs), derived from saturated and unsaturated fatty acids, have been identified and may exert similar biological effects by inhibiting the metabolic degradation of ECs. The ECL group also includes N-acylamino acids, N-acylneurotransmitters, and primary fatty acid amides. ECL compounds do not interact with cannabinoid receptors; they may exert cannabinoid-like effects by interacting with receptors such as GPR119 (G protein-coupled receptor 119), TRPV1 (Transient receptor potential vanilloid 1), and PPARα (Peroxisome proliferator-activated receptor α). EC and ECL compounds in humans are synthesized from dietary fatty acids; polyunsaturated n-6 and n-3 fatty acids constitute the most important sources. Studies have shown that changes in the type or amount of dietary fat can modulate EC and NAE levels in different tissues, which in turn can have an impact on health outcomes such as obesity and weight gain. EC and ECL compounds are found not only in humans but also in some plants and microorganisms. However, their profiles and concentrations in different food matrices are still poorly known, and there is limited information in the literature about the presence and formation of these compounds in foods. Based on this perspective, the aim of this thesis is to identify EC and ECL compounds in different fermented foods (sausage and cheese), to elucidate the formation and transformation mechanisms of these compounds, and to determine the biochemical changes they undergo during the digestive process. In addition, by evaluating the effects of these lipid-derived molecules, which play critical roles in the immune system, appetite regulation, pain transmission, and central nervous system functions, on the behavior of experimental animals consuming fermented foods with different EC/ECL profiles, this study seeks to clarify the potential impacts of fermented foods on food intake and diet-related mood. In the first part of the thesis, the EC/ECL compound profiles of different sausage samples were analyzed, revealing that these samples were rich in NAE, N-acylamino acids, and 2-MAG compounds. However, multiple factorial analysis (MFA) revealed no significant differences between fermented and heat-treated sausages with respect to processing conditions. The findings indicated that fermentation in laboratory-scale sausages increased the formation of N-acylamino acids and 2-MAG compounds. Conversely, heat treatment increased NAE levels and decreased the amount of 2-MAG. This decrease may be associated with the structural instability of 2-MAG compounds, which can lead to their conversion into 1-MAG isomers. However, the ripening/drying processes applied in laboratory-scale sausage production did not generally cause significant changes in EC and ECL compounds. During storage, a significant increase in N-acylamino acid concentrations was observed in fermented sausages, while 2-MAG levels were negatively affected. This chapter provides an important perspective on the effects of food processing methods on the profile and amounts of EC and ECL compounds in sausages. In the second part of the thesis, the EC/ECL compound profiles of fresh and ripened cheeses were compared, and it was determined that the ripening process is decisive in the formation of compounds involved in lipid signalling. Ripened cheeses, such as Danish Blue, were found to contain high levels of N-acylamino acid derivatives due to the intense lipolysis and proteolysis that occur during ripening. In cheddar cheese, N-oleoylphenylalanine, N-linoleoylphenylalanine, N-oleoylleucine, and N-palmitoylglycine levels increased during ripening. In contrast, the amount of 2-oleoylglycerol decreased significantly in fresh kashar cheese produced under laboratory conditions due to degradation during storage. Furthermore, it was concluded that 2-MAG compounds were found at higher concentrations in sausages compared to cheese samples due to their high fat content and the effects of lipolysis. The studies conducted in this part of the thesis provide insights into the effects of fermentation, ripening, and storage on the EC/ECL compound profile of foods and the possible mechanisms for their formation. In the third part of the thesis, model systems were designed to investigate the biosynthetic pathways and metabolic formation mechanisms of N-acylamino acid compounds in foods. In this context, findings obtained from a sausage-like model system indicated that microorganisms may play a role in the formation of N-acylamino acid compounds. This was further confirmed in the developed model medium system, where the formation of N-acylamino acid compounds (oleoylglycine and palmitoylglycine) was observed in the presence of amino acid (glycine), lipid, and starter culture, whereas no formation occurred in the absence of culture. These results suggest the involvement of a possible culture-derived N-acyltransferase-like enzyme. Furthermore, 2-MAG compounds were naturally present in the added oil, and their concentrations increased in the model system, consistent with sausage fermentation. However, 2-oleoylglycerol and 2-linoleoylglycerol were found to decrease in the presence of cultures in the medium. This difference is thought to result from the fact that only culture-derived enzymes are active in the medium model, whereas in the sausage-like model system, both microbial and muscle-derived endogenous lipolytic enzymes, together with the higher fat content, contribute to the process. The findings obtained from this section shed light on the possible mechanisms of endogenous or microbial enzymatic activities affecting the presence and formation of N-acylamino acid group compounds in foods. In the fourth part of the thesis, in vitro digestion experiments conducted on fermented sausage and aged kashar cheese demonstrated that these products were significant potential sources of EC and ECL compounds during the digestive process. An increase in N-acylamino acids and 2-MAG compounds was observed, particularly during the intestinal phase. It was determined that the increased levels of these compounds during digestion could be due to both their release from the structure and de novo synthesis processes. The findings of the in vitro digestion experiment conducted in the model system revealed that precursors (fatty acids and amino acids) are determinants of the formation of N-acylamino acid group compounds. Consequently, food composition and digestive enzymes appear to influence the profile of these bioactive lipids, shaping their potential health effects. The endocannabinoid system plays a critical role in appetite regulation and the control of energy balance. The studies conducted in the final part of the thesis demonstrated that different types of cheese could influence food intake and behavior in mice. Mice fed with fresh kashar cheese exhibited higher food consumption, which was associated with its EC/ECL profile, particularly the lower levels of 2-oleoylglycerol and N-acylamino acid compounds compared to aged kashar cheese. In contrast, the richer EC/ECL composition of aged kashar cheese was linked to appetite-suppressing effects, resulting in enhanced satiety and reduced anxiety-like behaviours in the mice fed this diet. These findings suggest that the EC/ECL profiles of cheeses may play a decisive role in energy balance and mood. In conclusion, this thesis study explores the effects of different processing methods on the EC/ECL compound profiles of fermented foods and the potential processes related to their formation mechanisms. It has been demonstrated that microorganisms play a critical role in the formation of N-acylamino acid compounds and that these compounds may be released during digestion. Additionally, it has been determined that consumption of ripened cheese may have effects on mood and appetite. These findings highlight that the composition and processing conditions of fermented foods may have potential impacts on health; however, further research is required to clearly elucidate the bioavailability and in vivo physiological effects of these lipid-derived compounds.
